摘 要:目的考察前胡汤对细菌脂多糖(LPS)诱导大鼠急性气道炎症和痰液分泌的影响及相关机制。方法大鼠采用气管注射LPS(300μg/500μl)建立模型,随机分为模型、急支糖浆、氨溴索口服液及前胡汤高、低剂量组(20、10ml/kg)。LPS注射前0.5h及造模后25h、49h、73h分别灌胃给药1次。末次给药2h后解剖取材,检测支气管肺泡灌洗液(BALF)白细胞分类计数、血清和BALF中炎症因子水平、肺组织病理学、肺组织黏蛋白(MUC5AC)水平及杯状细胞分化相关信号分子的表达。结果前胡汤可明显减少模型大鼠BALF中的白细胞数量且回复巨噬细胞、淋巴细胞等比例,显著降低血清和BALF中IL-4、5、9、13和TNF-α等炎症因子水平。肺组织病理学显示前胡汤明显改善大鼠肺泡结构和减轻炎细胞浸润。前胡汤可显著减少模型大鼠肺匀浆和BALF中MUC5AC黏蛋白水平,明显下调肺组织EGFR和STAT6的mRNA和蛋白表达,对SPDEF和NOTCH2表达水平作用不明显。结论前胡汤对LPS所致大鼠气道炎症及黏液分泌具有良好的改善作用,其抑制气道分泌作用与EGFR/STAT6信号通路介导的气道杯状细胞分化有关。Objective To investigate the influences of Qianhu(Peucedani Radix)decoction on bacterial lipopolysaccharide(LPS)’s inducing acute airway inflammation and sputum secretion in rats.Methods The rats were injected with LPS(300μg/500μl)to establish models and were divided randomly divided into model group,Jizhi Tangjiang group,ambroxol oral solution group,Qianhu(Peucedani Radix)decoction high and low dose group(20,10ml/kg).The rats were administered by gavage once 0.5h before LPS injection and 25h,49h and 73h after modeling respectively.Two hours after the last administration,the rats were dissected to detect the bronchoalveolar lavage fluid(BALF)’s differential blood count,the level of inflammatory factors in serum and BALF,lung histopathology,the level of mucin(MUC5AC)in lung tissue and goblet cell differentiation related signal molecules’expression.Results Qianhu(Peucedani Radix)decoction could significantly reduce the white blood cell quantity in BALF of model rats and restore the ratio of macrophages,lymphocytes and others,and remarkably reduce the levels of IL-4,5,9,13 and TNF-αand other inflammatory factors in serum and BALF.The lung histopathology shows that Qianhu(Peucedani Radix)decoction remarkably improves the alveolar structure of rats and reduces inflammatory cell infiltration.The Qianhu(Peucedani Radix)decoction could observably decrease the MUC5AC mucin level in lung homogenate and BALF of model rats,obviously lower the mRNA and protein expression of lung tissue EGFR and STAT6,but is not effective on the expression level of SPDEF and NOTCH2.Conclusion Qianhu(Peucedani Radix)decoction has a good improvement effect on the airway inflammation and mucus secretion of rats induced by LPS,and its inhibition on airway secretion is related to airway goblet cell differentiation mediated by EGFR/STAT6 signal pathway.
