摘 要:目的分别研究不同光强、采收时间、菌株等对灵芝多糖和三萜含量的影响,筛选适宜代料栽培灵芝的条件。方法采用紫外-可见分光光度法测定灵芝多糖和灵芝三萜含量,超高效液相色谱法测定灵芝子实体6种灵芝酸含量。结果研究发现,在320~400 lx光强条件下生长的灵芝子实体多糖含量最高,随着灵芝弹孢过程的持续,灵芝子实体的多糖、总三萜及6种灵芝酸总量均呈现下降趋势。通过对4种不同菌株栽培灵芝的质量评价,发现子实体和孢子粉的多糖含量高低依次为4号菌株>2号菌株>1号菌株>3号菌株,各菌株多糖含量差异显著(P<0.05);4号菌株子实体的灵芝酸总含量高达17.005mg/g,显著高于其他3个菌株(P<0.05)。结论研究结果显示4号菌株为适宜栽培的优良灵芝菌种,灵芝生长的光强条件应控制在320~400 lx,以子实体多糖为指标应该在弹孢前采收灵芝。Objective To study the effects of light intensity,harvesting time and strain on polysaccharides and triterpenoids in Ganoderma lucidum(G.lucidum)cultivated with substitute materials,so as to find the suitable conditions for G.lucidum cultivation.Methods The contents of G.lucidum polysaccharide(GLP)and G.lucidum triterpenoids(GLTs)were determined by UV-VIS.The contents of ganoderic acids(GAs)in fruiting bodies were determined by ultra high performance liquid chromatography(UPLC).Results The content of GLP in the fruiting bodies of G.lucidum grown under light intensity of 320-400 lx was the highest.With the continuous ejection of spores,the total contents of GLP,GLTs and six GAs in fruiting bodies of G.lucidum decreased.The quality evaluation of different four strains showed that the order of GLP content in the fruiting bodies and spores was strain 4>strain 2>strain 1>strain 3 with significant difference(P<0.05).The average content of GAs in fruiting body of strain 4 was 17.005mg/g,significantly higher than that of others(P<0.05).Conclusion The results showed that strain 4 was the dominant strain,and the light intensity of 320-400 lx was appropriate.Ganoderma lucidum should be harvested before sporulation based on fruiting body polysaccharide.
