摘  要:目的 探讨生地黄对过敏性紫癜(HSP)小鼠可能的作用机制研究。方法 将60只SPF级雄性实验小鼠,平均随机分成对照组、模型组、西咪替丁组(0.1 g/mL)、生地黄提取物低、中、高剂量组(10、20、30 mg/mL)。除对照组小鼠外,其余小鼠造模HSP,成功后按实验要求每日灌胃等量对应生地黄提取物,模型组和对照组灌胃等量生理盐水,连续21 d。结束后,采用碘酸雪夫染色肾脏组织的病理学变化,采用ELISA试剂盒检测小鼠血清IFN-γ、IL-2、IL-4、IL-10水平,Western blot法检测小鼠肾组织中p-JAK2(Janus Kinase 2,JAK2)、p-STAT3(signal transducer and activator of transcription 3,STAT3)的蛋白相对表达。结果 西咪替丁组和生地黄提取物组病理评分都明显远低于模型组,给药后小鼠肾脏组织学损害显著减轻。与对照组比较,模型组小鼠IFN-γ、IL-2表达均明显降低,IL-4、IL-10表达均明显升高(P <0.05);与模型组比较,西咪替丁组、生地黄提取物低、高剂量组小鼠IFN-γ、IL-2水平均升高,IL-4、IL-10水平均降低(P <0.05),且与生地黄提取物低、中剂量组相比,生地黄提取物高剂量组表达更明显(P <0.05)。与对照组相比,模型组小鼠肾组织中p-JAK2、p-STAT3蛋白表达均显著升高(P <0.01);生地黄提取物高剂量组小鼠肾组织中p-JAK2、p-STAT3蛋白表达明显低于西咪替丁组、生地黄提取物低、中剂量组(P <0.05)。结论 生地黄提取物具有显著的抗氧化活性和抗炎作用,可减轻HSP小鼠的症状,其作用机制可能与抑制JAK2/STAT3信号通路激活,从而调节p-JAK2和p-STAT3蛋白的表达水平有关。Objective To explore the possible mechanism of unprocessed rehmannia root on Henoch-Schönlein purpura(HSP)in mice.Methods Sixty SPF grade male experimental mice were randomly divided into a control group,a model group,a cimetidine group(0.1g/mL),and low-,medium-,and high-dose groups of unprocessed rehmannia root extract(10,20,30 mg/mL).Except for the mice in the control group,an HSP model was made in all the other mice.After that,they were treated by gavage with the corresponding experimental drugs of the same amount every day,while the model group and control group were gavaged with the same amount of normal saline,all for 21 consecutive days.At the end of the experiment,the pathological changes in renal tissue were measured by Schiff’s iodate staining,and the serum levels of IFN-γ,IL-2,IL-4 and IL-10 were measured by ELISA kits.Western blot was used to detect the relative protein expressions of p-JAK2 and p-STAT3 in the mouse kidney tissue.Results The pathological scores were significantly lower in both the cimetidine group and the unprocessed rehmannia root extract groups than those in the model group,and the histological damage to the kidneys of the mice was significantly reduced after administration of the drugs.Compared with those in the control group,the expressions of IFN-γand IL-2 were significantly decreased in the model group,while the expressions of IL-4 and IL-10 were significantly increased(P<0.05);Compared with those in the model group,the levels of IFN-γand IL-2 were increased in both the cimetidine group and the low-and high-dose groups of unprocessed rehmannia root extract,while the levels of IL-4 and IL-10 were decreased(P<0.05),and compared with those in the low-and medium-dose groups of unprocessed rehmannia root extract,the corresponding expression levels decreased or increased more significantly in the high-dose group(P<0.05).Compared with those in the control group,the expressions of p-JAK2 and p-STAT3 in the kidney tissue of mice were significantly increased in the model group(P<0.0
