薏苡附子败酱散对肠上皮细胞屏障损伤的保护作用研究

摘  要:目的 探讨薏苡附子败酱散对LPS诱导的肠上皮(IEC-6)细胞屏障损伤的保护作用。方法 IEC-6细胞分为对照组,模型组,薏苡附子败酱散全方组(以下简称全方组),全方去附子组(以下简称去附子组)和单用附子组。除对照组外,其余各组加入10μg/mL LPS(前期预实验确定该浓度为最佳造模浓度),模型组分别加入10 mg/mL全方、8.82 mg/mL全方去附子、1.18 mg/mL单味附子。MTT法检测各组细胞活性;ELISA法检测相关炎症因子含量;Western Blot法检测Occludin、ZO-1蛋白表达量;Transwell法检测细胞屏障通透性。结果 与对照组比较,模型组IEC-6活率显著下降(P <0.01),炎症因子TNF-α和IL-6含量明显增加(P <0.01),Occludin、ZO-1蛋白表达量显著降低(P <0.01),肠黏膜通透性显著增加(P <0.01)。全方组、去附子组均能提高LPS损伤下IEC-6细胞的活率(P <0.01),降低TNF-α和IL-6分泌量(P <0.01),上调ZO-1、Occludin的表达(P <0.01),降低跨膜电阻值(TEER)(P <0.01)。全方组在提高细胞活率作用强于去附子组(P <0.05),在降低TNF-α和IL-6分泌上2组作用相当(P> 0.05),在增加ZO-1蛋白表达上,全方组强于去附子组,而对Occludin的表达,2组作用相当(P> 0.05)。单味附子无明显抗炎作用(P> 0.05),但可提高损伤细胞的活性(P <0.05)。单味附子仅对ZO-1蛋白有明显促表达作用(P <0.01),但仍可明显降低细胞通透性(P <0.01)。结论在肠上皮细胞屏障损伤的保护作用上全方>去附子>单用附子,附子可能在全方中发挥了重要作用。Objective To investigate the protective effect of Yiyi Fuzi Baijiang Powder and its disassembled prescriptions on the intestinal epithelial(IEC-6)cell barrier injury induced by lipopolysaccharide(LPS).Methods IEC-6 cells were divided into a control group,a model group,a YFBS group,a YFBS removing Fuzi group(YFBSRF)and a Fuzi alone group(FZ).Except for the control group,10μg/mL LPS was applied to other groups,10mg/mL YFBS,8.82mg/mL YFBSRF,and 1.18mg/mL FZ were applied to the three administration groups,respectively.Cell activity of was detected by MTT assay.The content of inflammatory cytokines was detected by ELISA.The expression levels of tight junction proteins Occludin and ZO-1were detected by Western Blot.Cell barrier permeability were measured by Transwell.Results Compared with the indexes in the control group,the viability of IEC-6 was significantly decreased in the model group(P<0.01),the contents of TNF-αand IL-6 were significantly increased(P<0.01),the expression levels of Occludin and ZO-1 were significantly decreased(P<0.01),and the permeability of intestinal mucosa was significantly increased(P<0.01).Both the YFBS group and the YFBSRF group increased the viability of cells injured by LPS(P<0.01),decreased the secretion of TNF-αand IL-6(P<0.01),up-regulated the expression levels of ZO-1 and Occludin(P<0.01),and decreased TEER(P<0.01).The effect of the YFBS group in increasing cell viability was stronger than that of the YFBSRF group(P<0.05),and the effect of the YFBS group in decreasing TNF-αand IL-6 secretion was equal to that of the YFBSRF group(P>0.05).The effect of the YFBS group in increasing the protein expression of ZO-1 was stronger than that of the YFBSR group,while the effect in increasing the expression of Occludin was equal(P>0.05).Although the FZ group had no anti-inflammatory effect(P>0.05),it increased the activity of injured cells(P<0.05).Although the FZ group only promoted the protein expression of ZO-1 significantly(P<0.01),it still significantly reduced the cell permeability(P<0.
