摘　　要：刀豆蛋白A(concanavalin A,ConA)是一种来源于豆科植物种子的凝集素,一般以二聚体或四聚体形式存在,二聚体结构中主要含有β-折叠片和α-螺旋。作为一种金属蛋白,ConA的Mn^(2+)和Ca^(2+)配位域对其糖基结合域(carbohydrate-binding domain,CBD)的活性有重要作用。ConA能够特异性结合甘露糖和葡萄糖,因此利用ConA可特异性地识别并结合糖基且具有可逆性的特点,已被广泛应用于糖蛋白、糖脂等含糖物质和病毒的纯化。笔者综述了ConA的结构、理化特性、与糖基的相互作用及在含糖物质和病毒纯化方面的研究进展,并对ConA在兽医领域的应用前景进行了探讨,旨在为ConA的进一步开发利用提供参考。Concanavalin A(ConA)is a legume lectin derived from the seeds of the legume family,usually in the form of a dimer or tetrimer,with a dimer structure consisting mainly of aβ-folded sheet and anα-helix.As a metal protein,the Mn^(2+) and Ca^(2+) coordination domains of ConA play an important role in the activity of its carbohydrate-binding domain(CBD).ConA can specifically bind mannose and glucose,so it can specifically identify and bind glycogroups and has the characteristics of reversibility.ConA has been widely used in the purification of glycoproteins,glycolipids and other sugar-containing substances and viruses.The structure,physical and chemical properties,interaction with glycogroups and the research progress of ConA in the purification of sugar-containing substances and viruses were reviewed by the author,and the application prospect of ConA in the field of veterinary medicine was discussed.The purpose was to provide reference for the further developmentandutilizationof ConA.
