摘　　要：为分析牦牛PDK1基因的分子结构及理化特征,本研究通过RT-PCR扩增获得牦牛PDK1 CDS片段并进行克隆、测序和生物信息学初步分析,结果显示:牦牛PDK1基因CDS核苷酸大小为1137bp,编码氨基酸序列378个,具有较高保守性,其核苷酸序列与普通牛、人、野猪、大羚羊、绵羊、大熊猫、骆驼、长臂猿、大猩猩和山羊PDK1的核苷酸和氨基酸序列一致性分别高于9226%和9630%。生物信息学分析提示,牦牛PDK1基因编码蛋白属亲水蛋白,主要分布在细胞质和细胞核,其理论等电点769,分子量4333 KD,具有潜在磷酸化位点31个,也具有潜在N-糖基化、O-糖基化和甘露糖基化修饰位点。牦牛PDK1含BCDHK_Adom3和HATPase-PDK样保守区域,二级结构由α-螺旋、无规则卷曲和β-转角组成。本研究为进一步研究牦牛PDK1功能提供参考。To analyze the molecular structure and physicochemical characteristics of yak PDK1 gene,we cloned the yak PDK1 gene u-sing RT-PCR.By analyzing the cloned sequence of PDK1,we found PDK1 contained 378 amino acids,and PDK1 protein is 43.33 KD.The gene sequence identity and protein sequence similarity of PDK1 were higher than 92%in yak and other animals including Bos mutus,Bos taurus,Homo sapiens,Sus scrofa,Oryx dammah,Ovis aries,Ailuropoda melanoleuca,Camelus ferus,Hylobates moloch,Gorilla and Capra hircus.Our bioinformatic analysis indicated that the yak PDK1 has 31 potential phosphorylation sites,as well as other modification sites including N-glycosylation,O-glycosylation,and mannosylation.The second structure of yak PDK1 with both BCDHK_Adom3 and HATPase-PDK domains mainly includedα-helix,random coil andβ-sheet.Our results provide the basic data for further study on yak PDK1.
