摘 要:目的:观察加减地黄饮子对衰老小鼠睾丸组织沉默信息转录调节基因3(SIRT3)表达的影响。方法:选取2月龄雄性ICR小鼠作为青年组,20月龄小鼠作为老年组、维生素E组和加减地黄饮子组。青年组、老年组以生理盐水灌胃,维生素E组、加减地黄饮子组以对应药物灌胃。干预60 d后计算睾丸器官指数,观测睾丸组织病理学变化,检测各组小鼠血清睾酮和肝肾功能,检测睾丸组织丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)水平,以RT-PCR和Western-blot法检测SIRT3基因的mRNA和蛋白表达。结果:与老年组比较,加减地黄饮子组小鼠睾丸器官指数无差异(P>0.05);睾丸组织结构清晰稳定,精原细胞、精母细胞、支持细胞增多;血清睾酮水平升高,肝肾功指标正常,SOD、GSH-Px水平升高,MDA水平降低,SIRT3基因mRNA及其蛋白表达水平升高,差异均有统计学意义(P<0.05)。结论:加减地黄饮子可能影响SIRT3基因表达,减少小鼠睾丸氧化应激损伤而延缓睾丸衰老。Objective:To observe the effect of modified Dihuang Yinzi on the expression of silent information transcription regulatory 3(SIRT3)in testicular tissue of aging mice.Methods:Two-month-old male ICR mice were selected as Youth group,while 20-month-old mice were selected as Elderly group,Vitamin E group,and Modified Dihuang Yinzi group.Youth group and Elderly group were gavaged with physiological saline,while Vitamin E group and Modified Dihuang Yinzi group were gavaged with corresponding drugs.After 60 days of intervention,the testicular organ index was calculated,pathological changes in testicular tissue observed,serum testosterone and liver and kidney function measured in each group,levels of malondialdehyde(MDA),superoxide dismutase(SOD),and glutathione peroxidase(GSH-Px)in testicular tissue measured,and mRNA and protein expression of SIRT3 detected by RT-PCR and Western blot methods.Results:Compared with Elderly group,there was no significant difference in the testicular organ index of mice in Modified Dihuang Yinzi group(P>0.05);The testicular tissue structure was clear and stable,with an increase in spermatogonia,spermatocytes,and sertoli cells;Serum testosterone levels significantly increased,liver and kidney function indicators were normal,SOD and GSH-Px levels significantly increased,MDA significantly decreased,and mRNA and protein expression of SIRT3 significantly increased.The differences were statistically significant(P<0.05).Conclusion:Modified Dihuang Yinzi may affect the expression of SIRT3,reduce oxidative stress damage to mouse testes,and delay testicular aging.
