摘 要:目的探讨大黄酸对革兰阴性致病菌的体外抑菌作用及其机制。方法采用液体二倍稀释法测定大黄酸对大肠埃希菌、福氏志贺氏菌、鲍曼不动杆菌、肺炎克雷伯菌、绿脓杆菌、副溶血弧菌等革兰阴性致病菌的最低抑菌浓度(MIC)和最低杀菌浓度(MBC);同时以大肠埃希菌为对象,采用酶标仪检测核酸、蛋白质等细胞膜内含物的渗透量。结果大黄酸对大肠埃希菌、福氏志贺氏菌、鲍曼不动杆菌、肺炎克雷伯菌、绿脓杆菌、副溶血弧菌等标准菌株的MIC为0.31~5.00 mg/ml,MBC为0.63~10.00 mg/ml;同时在大黄酸作用后,大肠埃希菌细胞膜内的核酸、蛋白质外泄量明显增加。结论大黄酸对革兰阴性菌具有良好的抑菌作用,作用机制与破坏菌体细胞膜的通透性有关。Objective To investigate the antibacterial effect of rhein on gram-negative bacteria in vitro and its mechanism.Methods The MIC and MBC of rhein against gram-negative pathogenic bacteria such as Escherichia coli,Shigella flexneri,Acinetobacter baumannii,Klebsiella pneumoniae,Pseudomonas aeruginosa and Vibrio parahaemolyticus were determined by liquid double dilution method;At the same time,Escherichia coli was taken as the object,and the penetration amount of nucleic acid,protein and the other cell membrane contents were detected by microplate reader.Results The MIC of rhein against standard strains such as Escherichia coli,Shigella flexneri,Acinetobacter baumannii,Klebsiella pneumoniae,Pseudomonas aeruginosa,Vibrio parahaemolyticus was 0.31-5.00 mg/ml,and the MBC was 0.63-10.00 mg/ml;At the same time,the leakage of nucleic acid and protein in the cell membrane of Escherichia coli was significantly increased after rhein treatment.Conclusion Rhein has a good antibacterial effect on gram-negative bacteria,and its mechanism is related to the destruction of the permeability of cell membrane.
