摘 要:目的 通过观察天香丹对动脉粥样硬化(Atherosclerosis, AS)小鼠不对称二甲基精氨酸(Asymmetric dimethylarginine, ADMA)及沉默信息调节因子1(Sirtuin1,SIRT1)的影响,探讨天香丹对AS疾病血管内皮功能的调控作用。方法 采用ApoE^(-/-)小鼠(高脂饲料喂养)建立动脉粥样硬化模型,同时设C57BL/6J小鼠(普通饲料喂养)10只为空白组作为对照。造模成功后将ApoE^(-/-)小鼠按照随机数字表法分为模型组、他汀组、天香丹组,每组10只。天香丹组、他汀组分别给予天香丹2.7 g·kg^(-1)·d^(-1)、阿托伐他汀3 mg·kg^(-1)·d^(-1)灌胃,模型组给予0.5 mL蒸馏水灌胃,每日1次,连续8周。取小鼠血清用于检测总胆固醇(Total cholesterol, TC)、甘油三酯(Triglyceride, TG)、低密度脂蛋白(Low-density lipoprotein cholesterol, LDL-C)、高密度脂蛋白(High-density lipoprotein cholesterol, HDL-C)、一氧化氮(Nitrous oxide, NO)及ADMA含量,主动脉用于观察主动脉病理变化情况及检测SIRT1表达情况。结果 与空白组相比,模型组可见明显斑块形成及血管结构紊乱,斑块面积增加,脂质沉积明显,TC、TG、LDL-C、ADMA、SIRT1升高,HDL-C、NO下降,差异均有统计学意义(P<0.05);与模型组比较,他汀组可见少量斑块、内膜结构紊乱及少量内膜下脂质沉积,TC、TG、LDL-C降低,HDL-C、NO、SIRT1升高,天香丹组仅有少量斑块及内膜增厚,斑块面积及脂质沉积情况明显改善,TG、LDL-C、ADMA降低,HDL-C、NO、SIRT1升高,差异均有统计学意义(P<0.05);与他汀组对比,天香丹组NO升高,ADMA降低,差异有统计学意义(P<0.05)。结论 天香丹可以降低AS小鼠ADMA水平,提高SIRT1表达,升高NO水平以改善内皮细胞功能障碍抗动脉粥样硬化。Objective To investigate the impact of Tianxiangdan on asymmetrical dimethylarginine(ADMA)and sirtuin1(SIRT1)in mice with arteriosclerosis(AS)and to explore its therapeutic effects on vascular endothelial function of AS.Methods ApoE^(-/-)mice(high-fat feed feeding)were used to establish atherosclerotic models,and C57BL/6J mice(ordinary feed feeding)were blank group used as control group.Then modeling successful,10 ApoE^(-/-)mice in each group were randomly divided into model group,atorvastatin calcium group and Tianxiangdan group.The Tianxiangdan and atorvastatin calcium groups received gavage administration of Tianxiangdan(2.7 g·kg^(-1)·d^(-1))and atorvastatin calcium(3 mg·kg^(-1)·d^(-1))respectively,while the model groups were administered 0.5 mL of distilled water by gavage once daily for 8 weeks.Blood serum was collected for total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C),nitric oxide(NO)and ADMA level measurements.The aorta was examined for pathological changes and expression was detected of SIRT1.Results Compared with the blank group,the model group showed obvious plaque formation and vascular structure disorders,increased plaque area and lipid deposition,and increased TC,TG,LDL-C,ADMA and SIRT1,decreased HDL-C and NO,the differences were statistically significant(P<0.05).Compared with the model group,the atorvastatin calcium group had a small number of plaques and endometrial structural disorders,and a small amount of subintimal lipid deposition,and TC,TG,LDL-C were decreased,and HDL-C,NO,SIRT1 were increased,while the Tianxiangdan group only had a small amount of plaques and endometrial thickening,and the plaque area and lipid deposition were significantly improved,and TG,LDL-C,ADMA were decreased,HDL-C,NO,SIRT1 were increased,the differences were statistically significant(P<0.05).Compared with the atorvastatin calcium group,the NO was increased and the ADMA was decreased in Tianxiangdan group(P<0.05).Conclusion Tianxiangdan
