摘  要:目的探讨西北燥证对糖尿病大鼠水通道蛋白-1(Aquaporin-1,AQP1)、水通道蛋白-3(Aquaporin-3,AQP3)和CD4^(+)T、CD8^(+)T细胞表达的影响。方法选取16只正常大鼠,建立2型糖尿病(Type 2 diabetes mellitus,T2DM)大鼠模型,将造模成功的大鼠随机分为正常环境T2DM组、西北燥证T2DM组,每组8只。另选正常大鼠8只设为正常对照组。观察各组大鼠的一般情况,检测大鼠空腹血糖(Fasting blood glucose,FBG)、空腹血清胰岛素(Fasting serum insulin,FINS)、胰岛素抵抗指数(Homeostasis model assessment for insulin resistance,HOMA-IR)的变化,比较各组大鼠AQP1、AQP3、CD4^(+)T、CD8^(+)T细胞的表达水平,分析血清CD4^(+)、CD8^(+)与AQP1、AQP3表达水平的相关性。结果与正常对照组比较,正常环境T2DM组大鼠和西北燥证T2DM组大鼠FBG、FINS、HOMA-IR、AQP1、AQP3、CD4^(+)T、CD8^(+)T表达差异均有统计学意义(P<0.05);与正常环境T2DM组比较,西北燥证T2DM组大鼠FBG、FINS、HOMA-IR、AQP1、AQP3、CD4^(+)T、CD8^(+)T表达差异均有统计学意义(P<0.05)。结论西北燥证外环境可影响T2DM大鼠的血糖水平,其作用机制可能与调控AQP1、AQP3、CD4^(+)T、CD8^(+)T细胞表达有关。Objective To investigate the effect of Northwest dryness syndrome on the expression of aquaporin-1(AQP1),aquaporin-3(AQP),CD4^(+)T and CD8^(+)T cells in diabetic rats.Methods 16 rats were selected to establish the model of Type 2 diabetes mellitus(T2DM),and the successful rats were randomly divided into normal T2DM group and T2DM group with Northwest dryness syndrome,with 8 rats in each group.Another 8 normal rats were selected as normal control group.The general situation of the rats in each group were observed,the changes of fasting blood glucose(FBG),fasting serum insulin(FINS)and insulin resistance index(HOMA-IR)were detected,and the expression levels of AQP1,AQP3,CD4^(+)T and CD8^(+)T cells in rats in each group were compared,analyzed correlation between serum CD4^(+),CD8^(+)and the expression levels of AQP1,AQP3.Results Compared with normal control group,the expres-sions of FBG,FINS,HOMA-IR,AQP1,AQP3,CD4^(+)T and CD8^(+)T in T2DM group in normal environ-ment and T2DM group in Northwest dryness syndrome were significantly different(P<0.05).Compared with T2DM group in normal environment,the expressions of FBG,FINS,HOMA-IR,AQP1,AQP3,CD4^(+)T and CD8^(+)T in T2DM group with Northwest dryness syndrome were significantly different(P<0.05).Conclusion Northwest dryness syndrome can affect the blood glucose level in T2DM rats,and the mecha-nism of action may be related to regulating the expression of AQP1,AQP3,CD4^(+)T and CD8^(+)T cells.
