摘 要:为研究青贮廉价培养基对青贮微生物的影响情况,从贵州优质青贮全株玉米秸秆发酵饲料中采集样本,进行16S rRNA测序,通过对青贮微生物的分离培养,将复合青贮微生物菌剂分别接种到MRS肉汤培养基、乳清粉培养基、番茄汁培养基、玉米面培养基中37℃厌氧培养48 h,测定其pH变化、生长曲线、活菌数、菌剂产量等。研究表明:青贮玉米秸秆发酵微生物中魏斯氏菌属(Weissella)丰度值为51.2%,假单胞菌属(Pseudomonas)丰度值为34.25%,片球菌(Pediococcus)丰度值为12.75%;MRS肉汤培养基活菌数、吸光值(OD)、pH显著优于其他试验组(P≤0.05),乳清粉培养基发酵青贮微生物的OD值显著高于玉米面培养基组、番茄汁培养基组(P≤0.05)。综上所述,乳清粉培养基可替代MRS肉汤培养基作为更低成本的廉价培养基,进而推动青贮微生物添加剂的高效发展。To study the effect of cheap silage medium on silage microorganisms.In this study,samples were collected from Guizhou high quality whole silage corn straw fermented feed for 16S rRNA sequenc⁃ing.Through the isolation and culture of silage microorganisms,the complex silage microbial microbial agent was inoculated into MRS broth medium,whey powder medium,tomato juice medium and corn flour medium were incubated anaerobically at 37℃for 48 h,and pH change,growth curve,viable bacteria count,microbial yield and so on were measured.The results showed that the abundance of microorganisms in silage corn straw fermentation was mainly composed of Weissella(51.2%),Pseudomonas(34.25%)and Pediococcus(12.75%).The viable bacteria number,light absorption value(OD)and pH of MRS broth medi⁃um were significantly better than those of other groups(P≤0.05),and OD of silage microorganisms ferment⁃ed on whey powder medium was significantly higher than that in corn flour medium group and tomato juice medium group(P≤0.05).In conclusion,whey powder medium can replace MRS broth medium as a lowcost medium with lower cost,thus promoting the ef⁃ficient development of silage microbial additives.
