摘  要:甜菜根腐病是由多种真菌或细菌病原体引起的严重病害,给甜菜生产带来了严重的经济损失。本研究于2022年10月初以采自黑龙江大学呼兰校区的甜菜根腐病植株为试验样品,应用形态特征观察、致病性测定和分子生物学鉴定,结合科赫氏法则以及创建系统发育树和序列比对分析的方法,分离甜菜根腐病的致病菌。结果表明,本次呼兰校区腐烂的甜菜块根上分离得到可见白色菌丝菌株BY-1为镰孢属腐皮镰孢菌。小分生孢子通常是单细胞的,只有一个隔膜,但大分生孢子呈镰刀形,具有隔膜结构。分子鉴定结果显示,该病原菌在基于ITS和tef1序列构建的系统发育树上与腐皮镰孢菌类群处同一分支。基于形态学以及分子鉴定结果,发现导致该地区发生甜菜根腐病的病原菌是腐皮镰孢菌(Fusarium solani)。Sugar beet root rot is a severe disease caused by various fungal or bacterial pathogens,resulting in significant economic losses to sugar beet production.In this study,sugar beet with root rot harvested from Hulan campus of Heilongjiang University in early October 2022 were used as test samples to isolate the pathogens causing sugar beet root rot,using morphological characterization,pathogenicity determination and molecular biology identification,combined with Koch's rule and the methods of phylogenetic tree construction and sequence alignment analysis.The results showed that a visible white mycelial strain BY-1 was isolated from the rotted root of sugar beet taproots at the Hulan campus,identified as Fusarium spp.The large conidia were fusiform with septate structure,and the small conidia were usually unicellular with a small portion of septate.Molecular identification showed that the pathogen was in the same branch as the Fusarium spp.in the phylogenetic tree constructed based on ITS and tef1 sequences.Based on the morphological and molecular identification results,the pathogenic fungus causing the root rot of sugar beet in Hulan campus of Heilongjiang University was found to be Fusarium solani.
