分蘖洋葱伴生番茄根际微生物对番茄黄萎病抗性的影响

摘  要:为明确分蘖洋葱伴生番茄土壤微生物对减轻番茄黄萎病的作用,以分蘖洋葱伴生番茄为研究对象,通过植物-土壤微生物反馈试验分析分蘖洋葱伴生番茄根际微生物对黄萎病病情指数、发病率以及抗性相关酶及基因表达的影响。结果表明,在接种V.dahliae 10 d和15 d后,接种分蘖洋葱伴生番茄根际土处理(I)的番茄黄萎病发病率和病情指数显著低于接种单作番茄根际土处理(M)。在接种V.dahliae 48 h后,具有伴生根际土壤处理(TI)的番茄根系PR(PR 1、PR 2、PR 3)相对表达量显著高于具有单作根际土壤处理(TM),分别上调2.00倍、1.43倍和1.74倍。在接种V.dahliae 24 h、48 h和96 h后,TI的PPO相对表达量均显著高于TM,分别上调1.98倍、2.41倍和1.51倍;在接种V.dahliae 24 h和48 h后,TI处理的番茄根系PAL相对表达量均显著高于TM,分别上调1.20倍和1.19倍;与TM处理相比,TI处理的番茄根系LOX相对表达量在接种V.dahliae 96 h后显著上调1.37倍。在接种V.dahliae 48 h和96 h后,TI处理的PAL、PPO和β-1,3-葡聚糖酶活性显著高于TM处理;而TI处理SOD活性(229.06 U·g^(-1))仅在接种V.dahliae 48 h后显著高于TM处理。因此,分蘖洋葱伴生番茄减轻黄萎病病害,与分蘖洋葱伴生番茄根际土壤微生物提高番茄抗性基因表达和防御酶活性有关。To clarify the role of soil microbiota associated with potato onion in alleviating tomato wilt disease,we conducted a study using potato onion intercropped with tomatoes as our subject.Through plant-soil microbiota feedback experiments,we analyzed effect of rhizosphere microbiota of potato onion intercropped with tomatoes on the disease severity index,disease incidence,and the expression of disease resistance-related enzymes and genes in tomato wilt disease.The results showed that,10 and 15 days after inoculation with V.dahliae,the disease incidence and disease severity index of tomato wilt disease in the intercropping system with potato onion and tomato rhizosphere soil(I)were significantly lower than those in the monocropping system with tomato rhizosphere soil(M).48 hours after inoculation with V.dahliae,the relative expression levels of tomato root PR genes(PR1,PR2,PR3)in the intercropping rhizosphere soil treatment(TI)were significantly higher than those in the monocropping rhizosphere soil treatment(TM),with upregulations of 2.00 times,1.43 times,and 1.74 tims,respectively.At 24 hours,48 hours,and 96 hours after inoculation with V.dahliae,the relative expression of PPO in TI was significantly higher than in TM,with increases of 1.98 times,2.41 times,and 1.51 times,respectively.At 24 hours and 48 hours after V.dahliae inoculation,the relative expression of PAL in tomato roots of the TI treatment was significantly higher than that in the TM treatment,with increases of 1.98 times,2.41 tims,and 1.51 times,respectively.Compared to the TM treatment,the relative expression of LOX in tomato roots of the TI treatment significantly increased by 1.37 times at 96 hours after V.dahliae inoculation.At 48 hours and 96 hours after V.dahliae inoculation,the activities of PAL,PPO,andβ-1,3-glucanase in the TI treatment were significantly higher than in the TM treatment.However,the SOD activity in the TI treatment(229.06 U·g^(-1))was only significantly higher than in the TM treatment at 48 hours after V.dahliae inoculation.T
