摘 要:目的 以网络药理学为主要研究思路,探讨补肾化瘀方治疗宫腔粘连的潜在靶点与可能机制,以期为后续进一步临床及实验验证提供可靠的理论依据。方法 借助TCMSP、TCMID、PubChem、TCMSP、Swiss Target Prediction数据库对补肾化瘀方的活性成分、作用靶点进行筛选,借助Disgenet、Genecards、OMIM数据库获得宫腔粘连疾病靶点,将两者导入Cytoscape 3.8.2软件筛选交集靶点并构建“药物–成分–靶点–疾病”网络图,获得补肾化瘀方治疗宫腔粘连的核心成分,并通过STRING数据库构建的蛋白质相互作用(protein-protein interaction,PPI)网络图推测核心基因。最后通过R语言对交集靶点进行基因本体论(gene ontology,GO)与京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)富集分析,推测可能作用机制。结果 共获得补肾化瘀方治疗宫腔粘连的活性成分116个,起治疗作用的核心基因可能为CXCL8、KDR、LCK、ITGAL。GO富集至上皮细胞增殖、修复等生物学过程。KEGG富集分析共涉及169条信号通路,以涉及炎症、免疫激活的相关通路为主。结论 补肾化瘀方治疗宫腔粘连具有多靶点和多通路的复杂过程,基于网络药理学推测补肾化瘀方主要通过调控免疫–炎症反应、细胞增殖、转化等途径治疗宫腔粘连。Objective To explore the potential targets and mechanism of action of Bushen Huayu decoction in the treatment of intrauterine adhesion,based on network pharmacology in order to provide more theoretical basis for further clinical and experimental verification.Methods With the help of TCMSP,TCMID,PubChem,TCMSP and Swiss Target Prediction databases,the active components and targets of Bushen Huayu decoction were screened.With the help of Disgenet,Genecards and OMIM databases,the targets of intrauterine adhesion were obtained.The two components were imported into Cytoscape 3.8.2 software to screen the intersection targets and construct a ‘drug-component-target-disease’ network diagram to obtain the core components of the way that bushen huayu decoction in the treatment of intrauterine adhesion.The core genes of treatment were speculated by protein-protein interaction(PPI) network constructed by STRING database.Finally,gene ontology(GO) and Kyoto Encyclopedia of Genes and Genomes(KEGG) enrichment analysis of the intersection targets was performed by R language to speculate the possible mechanism of action.Results A total of 116 active components of Bushen Huayu decoction were obtained for the treatment of intrauterine adhesion.The core genes that play a therapeutic role may be CXCL8,KDR,LCK and ITGAL.GO enrichment to biological processes such as enzyme activity activation and epithelial cell proliferation repair.KEGG enrichment analysis involved 169 signaling pathways,mainly related to inflammation and immune activation.Conclusion The treatment of intrauterine adhesion with Bushen Huayu decoction has a complex process of multi-target and multi-channel.Based on network pharmacology,it is speculated that Bushen Huayu decoction mainly treats intrauterine adhesion by regulating immune-inflammatory response,cell proliferation and transformation.
