摘 要:目的:采用总评归一值结合响应面法优化黄连素水提取工艺。方法:以浸泡时间、料液比及每次煎煮时间为考察因素,以黄连素提取率及出膏率处理后总评归一(Overall Desirability,OD)值为响应值,采用Design-Expert 13.0软件,设计三因素、五水平试验。根据试验结果建立考察因素与响应值的数学模型,优化OD值最佳的提取工艺并进行验证。结果:在浸泡时间40 min、料液比13 kg/L、每次煎煮时间120 min条件下所得预测OD值最佳,为0.9474,经验证得黄连素平均提取率1.018%、平均出膏率10.98%、OD值0.9418,与预测值偏差-0.59%,所建立的数学模型预测性良好。结论:总评归一值结合响应面法优化黄连素最佳提取工艺条件稳定,考察指标在可控范围内最大,实际操作中具有一定可行性。Objective:To optimize the extraction process of berberine by overall desirability and response surface methodology.Methods:The optimal extraction conditions were determined based on the overall desirability(OD)of berberine extraction rate and paste yield,the soaking time,solid liquid ratio and time of each decoction were determined via central composite design-response surface methodology,using design-Expert 13.0 software to design three-factor,five-level tests.Aaccording to the test results,a mathematical model of the factors and response values was established,the optimal extraction process of OD value was optimized and verified.Results:When the soaking time was 40 min,the ratio of solid to liquid was 13 kg/L,and the decoction time was 120 min,the predicted OD value was the best,which was 0.9474.The average extraction rate of berberine was 1.018%,the average extraction rate was 10.98%,the OD value was 0.9418,and the deviation from the predicted value was-0.59%,the mathematical model was well predictive.Conclusion:The optimal extraction conditions of berberine by overall desirability and response surface methodology were stable,and was feasible in practice.
