摘 要:目的:本研究通过建立大鼠疼痛模型,从分子水平研究复方荆芥熏洗剂参与术后止痛的作用机制,完善对复方荆芥方止痛原理的认识,充分发挥中医药外治的优势,为临床治疗提供更多的实验理论依据,拓展临床应用前景。方法:实验室SPF级SD大鼠60只,雌雄各半,随机分为5组,分别为荆芥方组、抑制剂组、生理盐水组、模型组、空白组,每组12只。建立大鼠痔术后切口痛模型,荆芥方组给予荆芥方坐浴(1000 mL,每天2次,每次10 min),生理盐水组予0.9%氯化钠溶液坐浴,等量,每次10 min,抑制剂组予特异性拮抗剂普乐沙福(AMD3100)(1μg/10μL)外用。于给药的第3天取大鼠背根神经节,采用酶联免疫吸附法检测组织中炎性因子肿瘤坏死因子-α、白细胞介素-6表达水平变化;免疫荧光法检测大鼠背根神经节中CXCL12和CXCR4的表达水平,使用荧光显微镜观察复方荆芥熏洗剂对信号通路表达的影响。结果:与空白组相比,生理盐水组、模型组样本炎性因子肿瘤坏死因子-α、白细胞介素-6及信号通路CXCL12/CXCR4表达增高(P<0.05)。荆芥方组样本中炎性因子肿瘤坏死因子-α、白细胞介素-6及信号通路CXCL12/CXCR4的表达减少(P<0.05)。结论:荆芥方可能通过调控CXCL12/CXCR4信号通路表达,以及降低炎性因子肿瘤坏死因子-α、白细胞介素-6水平参与术后止痛。Objective:In this study,by establishing a rat pain model,the mechanism of action of the Jingjie prescription lotion participating(荆芥熏洗剂)in postoperative pain relief was studied from the molecular level,so as to improve the understanding of the principle of c the Jingjie prescription,give full play to the advantages of external treatment of TCM medicine,provide more experimental theoretical basis for clinical treatment,and expand the clinical application prospects.Methods:Sixty SD rats,half male and half female,were randomly divided into 5 groups,12 rats in each group,including the Jingjie prescription group,the inhibitor group,the physiological saline group,the model group and the blank group.The rat model of wound pain after hemorrhoids operation was established.The Jingjie prescription group was given Jingjie prescription sitbath(1000 mL,twice a day,for 10 minutes each time),the physiological saline group was given sitbath with normal physiological saline for 10 minutes each,and the inhibitor group was given specific antagonist AMD3100(1μg/10μL)for external use.On the third day of administration,the dorsal root ganglia of rats were taken,and the expressions of CXCL12 and CXCR4 were detected by immunofluorescence technique.The effect of compound Jingjie prescription fumigant on the expression of signal pathway was observed by fluorescence microscope.Results:Compared with the blank group,the expression of TNF-α,IL-6,CXCL12/CXCR4 in the physiological saline group and the model group were increased(P<0.05).The expression of TNF-α,IL-6,CXCL12/CXCR4 in the Jingjie prescription group were decreased(P<0.05).Conclusion:The Jingjie prescription may be involved in postoperative analgesia by regulating the expression of CXCL12/CXCR4 signaling pathway and reducing the levels of TNF-αand IL-6.
