摘  要:目的:运用网络药理学研究益气温阳护卫汤治疗哮喘的分子机制。方法:通过中药系统药理学数据库与分析平台(TCMSP)检索益气温阳护卫汤中十味中药的主要化学成分,通过TCMSP、DrugBank、Swiss平台查找及预测中药化合物对应靶点,在OMIM、Malacards、CTD、Disgent数据库中搜索哮喘相关靶点,取二者交集导入Swiss平台,获得蛋白质-蛋白质相互作用网络,筛选核心作用靶点并构建益气温阳护卫汤-潜在靶点-哮喘作用网络。对核心靶点进行基因本体论(GO)分子功能及京都基因与基因组百科全书(KEGG)富集分析。采用DiscoverStiudio软件对活性成分与靶点进行分子对接。结果:筛选得益气温阳护卫汤中主要活性成分221个,该活性成分有效靶点为770个,哮喘疾病靶点259个,取交集得核心靶点66个。GO富集分析显示靶点基因主要涉及炎症反应、凋亡过程的负调控、调控血管生成、酶结合、细胞因子活性、血红素结合、激酶活性等;KEGG信号通路主要包括癌症通路、缺氧诱导因子-1信号通路、肿瘤坏死因子(Tumor Necrosis Factor,TNF)信号通路、钙信号通路、核因子-κB信号通路等;分子对接结果中益气温阳护卫汤中主要活性成分与哮喘疾病靶点对接成功。结论:益气温阳护卫汤可通过调控TNF信号通路、核因子-κB信号通路、炎症介质对瞬时受体电位(Transient Receptor Potential,TRP)通道的调节,参与促炎症细胞因子的转录调节、过敏原诱导的炎症和气道上皮重塑等治疗哮喘。Objective:To study molecular mechanism of the Yiqi Wenyang Huwei decoction(益气温阳护卫汤)on asthma bsaed on network pharmacology.Methods:TCMSP database was used to search the main chemical components of ten TCM medicine in the Yiqi Wenyang Huwei decoction,and TCMSP,DrugBank and Swiss were used to search and predict the corresponding targets of TCM medicine.OMIM,Malacards,CTD and Disgent databases were used to search for asthma-related targets,and the intersection of the two targets was imported into Swiss to obtain PPI network.The core targets were screened and the network of the Yiqi Wenyang Huwei decoction-potential targets-asthma was constructed.The molecular function of GO and enrichment analysis of KEGG were carried out on the core targets.DiscoverStiudio software was used to carry out molecular docking between active ingredients and targets.Results:After screening,there were 221 main active components in the Yiqi Wenyang Huwei decoction,and the effective targets of this active component were 770,with 259 disease targets of asthma.Sixty-six core targets were obtained by intersection.GO enrichment analysis showed that the target genes mainly involved in inflammatory reaction,negative regulation of apoptosis,regulation of angiogenesis,enzyme binding,cytokine activity,heme binding,kinase activity,etc..KEGG signaling pathway mainly included cancer pathway,HIF-1 signaling pathway,tumor necrosis factor signaling pathway,calcium signaling pathway,NF-κB signaling pathway,etc..In the molecular docking results,the main active ingredients in the Yiqi Wenyang Huwei decoction successfully docked with asthma disease targets.Conclusion:The Yiqi Wenyang Huwei decoction can regulate tumor necrosis factor signaling pathway,NF-κB signaling pathway and regulation of transient receptor potential channels by inflammatory mediators.It is involved in the transcription regulation of pro-inflammatory cytokines,allergen-induced inflammation and airway epithelial remodeling in the treatment of asthma.
