摘 要:目的探讨芹菜素逆转毒胡萝卜素(TG)所致人神经母细胞瘤(SH-SY5Y)细胞内质网应激诱导凋亡的分子机制。方法采用细胞计数试剂-8(CCK-8)检测不同浓度TG和不同处理时间对SH-SY5Y细胞活力的影响,观察芹菜素对SH-SY5Y细胞的保护作用;蛋白免疫印迹(Western blotting)法检测SH-SY5Y细胞内质网应激球蛋白结合蛋白(BIP)、C/EBP同源蛋白(CHOP)、半胱氨酸天冬氨酸蛋白水解酶12(CASPASE-12)及细胞凋亡Bcl-2相关X蛋白(Bax)、B细胞淋巴瘤-2蛋白(Bcl-2)表达变化。结果与对照组(CON组)比较,TG可浓度和时间依耐性降低SH-SY5Y细胞活力(P<0.01);10、20及30μmol·L^(-1)芹菜素处理SH-SY5Y细胞1 h,与1.0μmol·L^(-1)TG比较,10μmol·L^(-1)芹菜素能逆转细胞活力(P<0.01),效果优于20、30μmol·L^(-1)芹菜素;芹菜素预处理能降低BIP、CHOP表达(P<0.05),增强Bcl-2表达(P<0.05),升高Bcl-2/Bax值(P<0.05)。结论芹菜素能够逆转TG诱导的SH-SY5Y内质网应激,增强细胞活力,抑制内质网应激所致细胞凋亡。Objective To investigate the effects of apigenin(Api)on human neuroblastoma(SH-SY5Y)cells,and to clarify the molecular mechanisms of Api on thapsigargin(TG)-induced endoplasmic reticulum stress and apoptosis.Methods CCK-8 was used to detect the effects of different concentrations of TG and different treatment times on SH-SY5Y cell viability and to observe the protective effect of Apigenin on SH-SY5Y cells.The protein expression of Endoplasmic reticulum stress globulin binding protein(BIP),Cyclic adenosine monophosphate response element binding transcription factor homolog(CHOP),Aspartate protein hydrolase of cysteine 12(CASPASE-12),Apoptosis-related proteins Bax(Bcl-2 Associated X Protein)and Bcl-2(B cell lymphoma-2)were examined using Western blotting.Results Compared with the control group,TG could reduce SH-SY5Y cell viability(P<0.01)depending on concentration and time.Compared with the 1.0μmol·L^(-1)TG group,Api could reverse cell viability after treated for 1 h(P<0.01),and the effect in 10μmol·L^(-1)group was better than those in 20 and 30μmol·L^(-1)groups.At the same time,Api pretreatment could reduce BIP and CHOP expression(P<0.05),increased the expression of the apoptosis marker Bcl-2(P<0.05),and increased the Bcl-2/Bax ratio(P<0.05).Conclusion Api treatment reverses endoplasmic reticulum stress induced by TG,increases cell viability,and inhibits endoplasmic reticulum stress-induced apoptosis.
