摘 要:目的基于中药提取物和含药血清两个层次研究左氧氟沙星(LEV)联合芪归银方(QGYD)对铜绿假单胞菌标准菌株及耐药菌株生物膜和绿脓菌素的影响,为中药联合抗菌药物防治生物膜感染的临床用药提供参考。方法制备药液(LEV组、LEV+QGYD组)和LEV组、LEV+QGYD组系列含药血清,分别干预铜绿假单胞菌标准菌株和临床多重耐药菌株,测定铜绿假单胞菌生长量、生物膜形成量和绿脓菌素分泌量。测定LEV组、LEV+QGYD组含药血清中LEV的含量和各组大鼠血清中TNF-α和IL-2的水平。结果LEV组和LEV+QGYD组(药液/血清)均能抑制铜绿假单胞菌(标准/耐药)的生长、生物膜的形成和绿脓菌素的分泌;LEV组与LEV+QGYD组血清中LEV含量分别为6.14μg·mL^(-1)和5.12μg·mL^(-1)。与空白对照组相比,LEV组TNF-α表达水平明显升高,IL-2表达水平无明显变化;QGYD组、QGYD+LEV组TNF-α和IL-2均无明显变化。与LEV组相比,QGYD+LEV组TNF-α和IL-2表达水平均明显降低。结论体外和半体内研究均表明,联用QGYD能增强LEV的抗菌活性,有效抑制铜绿假单胞菌生物膜的形成和绿脓菌素的分泌。Objective To determine the effect of levofloxacin(LEV)combined with Qiguiyin decoction(QGYD)on the biofilm and pyocyanin in Pseudomonas aeruginosa of standard and drugresistant strains,and to provide reference for the treatment of biofilm infection with traditional Chinese medicine combined with antibiotics.Methods The solution(an LEV group and an LEV+QGYD group)and series of medicated serum of the LEV group,and the LEV+QGYD group were prepared to interfere with the standard strains of Pseudomonas aeruginosa and multi-drug resistant strains,respectively.The growth amount of Pseudomonas aeruginosa,the formation biofilm and the secretion pyocyanin was measured.The content of the LEV in LEV group and the LEV+QGYD group and the levels of TNF-αand IL-2 in the serum of each group were determined.Results The LEV group and LEV+QGYD group(liquid/serum)inhibited the growth of Pseudomonas aeruginosa(standard/drug-resistant),the formation of biofilm and the secretion of pyocyanin.The serum LEV content in the LEV group and the LEV+QGYD group was 6.14μg·mL^(-1)and 5.12μg·mL^(-1),respectively.Compared with the control group,the expression level of TNF-αwas much increased in the LEV group,and the expression level of IL-2 was not obviously changed.Compared with the control group,there was no significant change in TNF-αand IL-2 in the QGYD group and the QGYD+LEV group.Compared with the LEV group,the expression levels of TNF-αand IL-2 in the QGYD+LEV group were greatly decreased.Conclusion The antibacterial activity of LEV is enhanced when combining with QGYD,and the formation of biofilm and secretion of pyocyanin in pseudomonas aeruginosa are effectively inhibited.
