摘 要:目的 探究肉桂醛(CA)对慢性阻塞性肺疾病(COPD)大鼠气道炎症的影响以及对SDF-1/CXCR4轴的调节机制。方法 将Wistar雄性大鼠随机分为对照组、模型组、氨茶碱组(阳性药物)、AMD3100组(CXCR4抑制剂)、CA低剂量组、CA高剂量组,每组10只。利用动物肺功能检测仪检测大鼠肺功能;HE染色观察肺组织病理学变化;收集支气管肺泡灌洗液(BALF),进行炎性细胞计数;ELISA检测BALF中炎症因子白三烯B4(LTB4)、白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)的水平;Western Blot检测肺组织SDF-1、CXCR4蛋白表达。结果 与对照组相比,模型组大鼠肺泡间隔变窄并断裂,肺泡扩大成肺大疱,支气管道周围明显炎症细胞浸润,FRC值、BALF中白细胞、中性粒细胞和淋巴细胞数量、促炎因子LTB4、IL-6、IL-1β、TNF-α的含量及SDF-1、CXCR4蛋白表达均升高,FEV_(0.3)/FVC及PEF值降低(P<0.05);与模型组相比,氨茶碱组和CA组大鼠肺泡结构恢复,肺泡轻微扩张,炎症细胞浸润减少,FRC值、BALF中白细胞、中性粒细胞和淋巴细胞数量、促炎因子LTB4、IL-6、IL-1β、TNF-α的含量及SDF-1、CXCR4蛋白表达均降低,FEV_(0.3)/FVC及PEF值升高(P<0.05)。结论 CA可能通过抑制SDF-1/CXCR4轴来减轻COPD大鼠的气道炎症。Objective To explore the impacts of cinnamaldehyde(CA)on airway inflammation in rats with chronic obstructive pulmonary disease(COPD)and its regulatory mechanism on the SDF-1/CXCR4 axis.Methods Wistar male rats were randomly grouped into a control group,model group,aminophylline group(positive drug),AMD3100 group (CXCR4 inhibitor), CA low dose group, and CA high dose group, with 10 rats in each group. An animal lung function detector was applied to detect rat lung function;HE staining was applied to observe the pathological changes of lung tissue;bronchoalveolar lavage fluid (BALF) was collected and inflammatory cells were counted;ELISA was applied to detect the levels of inflammatory factors such as leukotriene B4 (LTB4), interleukin-6 (IL-6), interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) in BALF;Western blot was applied to detect the expression of SDF-1 and CXCR4 proteins in lung tissue. Results Compared with the control group, the alveolar septa of rats in the model group narrowed and ruptured, the alveoli expanded into bullae, and inflammatory cells infiltrated around the bronchi, the FRC value, the numbers of white blood cells, neutrophils, and lymphocytes in BALF, the contents of pro-inflammatory factors LTB4, IL-6, IL-1β, TNF-α, and the expression of SDF-1 and CXCR4 proteins increased, the values of FEV 0.3 /FVC and PEF decreased ( P <0.05);Compared with the model group, the alveolar structure of rats in aminophylline group and CA group recovered, the alveoli expanded slightly, and the infiltration of inflammatory cells decreased, the FRC value, the numbers of white blood cells, neutrophils and lymphocytes in BALF, the contents of pro-inflammatory factors LTB4, IL-6, IL-1β, TNF-α, and the expression of SDF-1 and CXCR4 proteins decreased, the values of FEV 0.3 /FVC and PEF increased ( P <0.05). Conclusion CA may alleviate the airway inflammation of COPD rats by inhibiting the SDF-1/CXCR4 axis.
