摘 要:目的:观察三生调脂舒对糖尿病血糖波动大鼠脂蛋白磷脂酶A2(Lp-PLA2)活性表达的影响。方法:将90只大鼠分为正常组(Con组)、持续糖尿病高血糖组(Model-1组)、糖尿病血糖波动对照组(Model-2组)、三生调脂舒低剂量组(SSTZS-L组)、三生调脂舒中剂量组(SSTZS-M组)和三生调脂舒高剂量组(SSTZS-H组)。糖尿病血糖波动模型构建成功后,采用三生调脂舒干预,观察其对葡萄糖曲线下面积(AUG)、平均血糖波动幅度(MAGE)、三酰甘油(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL-C)及炎性因子白细胞介素-1α(IL-1α)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)及氧化和抗氧化指标丙二醛(MDA)、超氧化物歧化酶(SOD)、C肽及血清和腹主动脉Lp-PLA2含量变化。结果:与Con组比较,5个时间点(0、30、60、120、180 min)Model-1组、Model-2组及SSTZS各剂量组血糖升高(P<0.05),SSTZS各剂量组低于Model-1组和Model-2组(P<0.05)。与Con组比较,Model-1组、Model-2组及SSTZS各剂量组MAGE、AUG、TC、TG、LDL-C、IL-1α、IL-6、TNF-α和MDA升高(P<0.05),HDL-C、SOD和C肽降低(P<0.05);且SSTZS各剂量组MAGE、AUG、TC、TG、LDL-C、IL-1α、IL-6、TNF-α和MDA低于Model-1组和Model-2组(P<0.05),HDL-C、SOD和C肽高于Model-1组和Model-2组(P<0.05)。Con组细胞排列整齐规则,血管壁纤维呈明显规则状,Model-1组和Model-2组细胞核排列紊乱,与Model-1组和Model-2组相比,SSTZS各剂量组明显好转。与Con组比较,Model-1组、Model-2组及SSTZS各剂量组血清和腹主动脉中Lp-PLA2表达升高(P<0.05),SSTZS各剂量组低于Model-1组和Model-2组(P<0.05)。结论:三生调脂舒可调节Lp-PLA2表达和血脂,降低炎症反应,提高抗氧化能力,改善糖尿病大鼠血糖波动。Objective:To observe effects of Sansheng Tiaozhishu(SSTZS)on the expression of lipoprotein phospholipase A2(Lp-PLA2)activity in diabetic rats with blood sugar fluctuation.Methods:Ninety rats were divided into normal group(Con group),persistent diabetic hyperglycemia group(Model-1 group),diabetic blood glucose fluctuation control group(Model-2 group),Sansheng Tiaozhishu low-dose group(SSTZS-L group),Sansheng Tiaozhishu medium-dose group(SSTZS-M group),and Sansheng Tiaozhishu high-dose group(SSTZS-H group).After the diabetes blood sugar fluctuation model was constructed,Sansheng Tiaozhishu was used to intervene treatment.The changes of area under curve of glucose(AUG),mean amplitude of glycemic excursions(MAGE),lipid triacylglycerol(TG),total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C)and low-density lipoprotein cholesterol(LDL-C),the inflammatory factors interleukin-1α(IL-1α),interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),oxidative and antioxidant indexes malondialdehyde(MDA),superoxide dismutase(SOD),C peptide,and the content of serum and abdominal aorta Lp-PLA2 were observed.Results:Compared with the Con group,blood glucose in the Model-1 group,Model-2 group,and SSTZS dose groups was increased at 5 time points(0,30,60,120,and 180 min)(P<0.05),and blood glucose in the SSTZS dose groups was lower than that in the Model-1 group and Model-2 group(P<0.05).Compared with the Con group,MAGE,AUG,TC,TG,LDL-C,IL-1α,IL-6,TNF-α,and MDA in the Model-1 group,Model-2 group,and SSTZS dose groups were higher,and HDL-C,SOD,and C-peptide were lower(P<0.05).Compared with the Model-1 group and Model-2 group,MAGE,AUG,TC,TG,LDL-C,IL-1α,IL-6,TNF-α,and MDA in the SSTZS dose groups were lower,and HDL-C,SOD,and C-peptide were higher(P<0.05).The cells in the Con group were neatly and regularly arranged,and the vascular wall fibers were distinctly regular,while the nuclei of the cells in the Model-1 group and Model-2 group disorganized.Compared with the Model-1 group and Model-2 group,in SSTZS dose groups there we
