摘 要:明确在福建省武夷山市艳锦竹芋上发现的叶斑病病原菌,以期为该病害的防控提供参考。2021年采集武夷学院园艺大棚内患叶斑病的艳锦竹芋(Stromanthe sanguinea‘Triostar’)叶片,通过传统微生物培养方法分离纯化病原菌,并按照科赫氏法则测定其致病性,并对其进行形态学鉴定和基于ITS序列的分子鉴定,并通过系统发育树的构建,分析其遗传多样性。结果表明:从病叶中分离得到病原菌(标记为YJ-2),具有致病性,且与田间发病症状一致。基于ITS序列进行PCR扩增和测序,序列表明菌株YJ-2和NCBI核酸数据库中拟茎点霉属菌Phomopsis sp.(登录号DQ780431.1)的相似性为99.43%,且在系统发育树上聚在一起。根据形态特征和分子鉴定结果,将该地区艳锦竹芋叶斑病病原菌鉴定为拟茎点霉属菌,为国内首次报道拟茎点霉属菌引起的艳锦竹芋叶斑病。The pathogen causing leaf spot on Stromanthe sanguinea that found in Wuyishan city,Fujian province was confirmed so as to provide reference for the prevention and control of this disease.The leaves of S.sanguinea with spot disease were collected from gardening greenhouses of Wuyi University on 2021.The pathogens were isolated and purified by traditional microbial culture method and the pathogenicity was verified by Koch's rule.The pathogen specie was identified using morphological method and ITS-based molecular method.Moreover,the phylogenetic tree was constructed to analyze the fungus genetic diversity.Results showed that the pathogen(named YJ-2)isolated from diseased leaves were pathogenic to the host plant and the symptom was consistent with that in field.PCR amplification and sequencing were conducted based on ITS sequence.The results showed that the strain YJ-2 was 99.43%similar to Phomopsis sp.(Accession number DQ780431.1)in NCBI nucleic acid database,and gathered together on the phylogenetic tree.Based on the morphological features and molecular identification results,the pathogen of S.sanguinea leaf spot was identified as Phomopsis sp.,which was the first report of Phomopsis sp.causing S.sanguinea leaf spot in China.
