摘  要:目的:采用高效液相色谱法测定达原饮中槟榔生物碱的含量。方法:色谱条件为:Venusil SCX色谱柱(4.6 mm×250 mm,5μm,300Å),流动相:甲醇:0.2%磷酸(氨水调节pH值至3.8)=60:40,检测波长:210 nm,柱温:30℃,流速:1.0 mL•min^(-1)。结果:4种槟榔生物碱分离度良好,各组分在浓度范围内线性关系良好,加标回收率(n=9)在98.37%~102.11%,RSD值均小于2%。达原饮药液中去甲槟榔碱、槟榔碱、去甲槟榔次碱和槟榔次碱的含量分别为0.014 mg•mL^(-1)、0.046 mg•mL^(-1)、0.036 mg•mL^(-1)和0.018 mg•mL^(-1)。结论:该方法准确度高、稳定性强、灵敏度和重复性良好,简便可靠,可用于达原饮中槟榔生物碱的含量测定及质量控制。Objective:A method for the determination of areca alkaloids in Dayuanyin was established by HPLC.Methods The determination was performed on A Venusil SCX column(4.6 mm×250 mm,5µm,300 A)with mobile phase consisting of methanol:0.2%phosphoric acid(pH adjusted to 3.8 by ammonia)=60:40.Detection wavelength was set at 210 nm,column temperature at 30℃,and flow rate at 1.0 mL·min^(-1).Results The four areca alkaloids were well separated,and the linear relationship of each component was good in the concentration range.The standard recoveries(n=9)ranged from 98.37%to 102.11%,and RSD were all less than 2%.The contents of guvacoline,arecoline,guvacine hydrochloride and arecaine in Dayuan decoction were 0.014 mg·mL^(-1),0.046 mg·mL^(-1),0.036 mg•mL^(-1)and 0.018 mg·mL^(-1),respectively.Conclusion The method has high accuracy,stability,sensitivity and repeatability,is simple and reliable,and can be used for the content determination and quality control of areca alkaloids in Dayuan drink.
