摘  要:【目的】围绕内江猪精液品质和冷冻精液流程优化进行研究,以更好实现对内江猪的保种和开发利用。【方法】使用同一时期采精的5头内江猪精液检测了精子的活率、活力、运动状态和生化指标(甘油三酯、过氧化氢等),分析精子活力和精浆各生化指标相关性,并根据课题组此前的研究,进行了冷冻精液流程优化。【结果】平衡12 h后内江猪精液活力指标显著下降,原地移动精子数显著上升(P<0.05)。精子活力与精浆H_(2)O_(2)和MDA含量呈负相关,而与TG含量呈正相关关系。采用温和的离心方式(2000×g,7 min)、较高的甘油浓度(终浓度8%)对冷冻精液流程优化后,精子冷冻前活率提升了10.28%,活力提升了15.75%(P<0.05)。在低温慢速(37℃,20 s)条件下,冷冻保存内江猪精液解冻后活率最高(53.18±5.68)%。【结论】根据内江猪精液特点,优化后的冷冻程序和低温慢速(37℃,20 s)的解冻程序更适合内江猪。【Objective】This study was conducted to make a better realize of the conservation,develop⁃ment and utilization of Neijiang pigs,this study focused on the semen quality of Neijiang pigs and the optimization of the frozen semen process of Neijiang pigs.【Method】The semen of 5 Neijiang pigs collected during the same period was used to detect the sperm motility,motility,motility and biochemical indica⁃tors(triglyceride,hydrogen peroxide,etc.),and the correlation between sperm motility and seminal plasma biochemical indicators was analyzed.,and optimized the frozen semen process according to the previous research of the research group.【Result】After 12 hours of equilibration,the semen motility index of Neijiang pigs decreased significantly,and the number of sperm that moved in situ increased sig⁃nificantly(P<0.05).Sperm motility was negatively correlated with seminal plasma H_(2)O_(2) and MDA con⁃tent,but positively correlated with TG content.After optimizing the frozen semen process by using mild centrifugation(2000×g,7 min)and high glycerol concentration(final concentration 8%),the sperm motility rate before freezing was increased by 10.28%,and the motility was increased by 15.75%(P<0.05).Under the condition of low temperature and slow speed(37℃,20 s),the survival rate of frozen preserved Neijiang pig semen after thawing was the highest(53.18±5.68)%.【Conclusion】According to the characteristics of Neijiang pig semen,the optimized freezing procedure and low temperature slow(37℃,20 s)thawing procedure are more suitable for Neijiang pigs.
