摘  要:奶牛产后子宫内膜炎可导致子宫复旧延迟,从而导致繁殖性能下降。试验旨在探讨益母草碱对产后子宫复旧不全的影响以及细胞凋亡相关基因及蛋白的表达情况,为益母草碱治疗母畜产后子宫复旧不全提供新的思路和理论支持。研究通过对60只产后小鼠子宫灌注LPS建立子宫复旧不全模型,并分别给予不同浓度的益母草碱(0.5,1.5,4.5 mg/kg)处理1或3 d。病理切片发现,LPS能导致子宫内膜上皮的延迟更新、子宫腺扩张,影响血流状态等,出现一定程度的子宫复旧不全;益母草碱治疗后,充血、出血减轻,炎性细胞浸润减少,且呈剂量依赖性。Real-time PCR法检测细胞凋亡相关基因Bax和Bcl-2的表达,研究发现模型组Bax表达上升,益母草碱处理后表达下降,且呈剂量依赖性;模型组Bcl-2表达下降,益母草碱处理后表达上升。免疫印迹检测相关蛋白表达水平变化,发现模型组Bax表达增加、Bcl-2表达减少,益母草碱处理后Bax表达减少,Bcl-2表达增加。结果表明,益母草碱通过对Bax、Bcl-2的调控,抑制子宫内膜细胞的凋亡,促进产后子宫复旧。Medicine,Huazhong Agricultural University;Postpartum endometritis in dairy cows can cause delayed involution of the uterus,resulting in decreased reproductive performance.The aim of study was to investigate the effect of leonurine on postpartum uterine subinvolution and the expression of apoptosis-related genes and proteins,and to provide new ideas and theoretical support for the treatment of postpartum uterine subinvolution with leonurine.60 postpartum mice were perfused with LPS to establish the model of uterine subinvolution and treated with different concentrations of leonurine(0.5,1.5,4.5 mg/kg)for one or three days in study,.The pathological sections showed that LPS can cause delayed endometrial epithelial renewal,uterine gland expansion,affect the state of blood flow,etc.,and a certain degree of uterine subinvolution;the hyperemia and hemorrhage were alleviated,and inflammatory cell infiltration was reduced in a dose-dependent manner after treatment with leonurine.Real-time PCR was used to detect the expression of apoptosis-related genes Bax and Bcl-2,and it was found that the expression of Bax increased in the model group,but decreased in a dose-dependent manner after treatment with leonurine;the expression of Bcl-2 in model group was decreased,the expression of Bcl-2 increased after treatment with leonurine.Western blot showed that the expression of Bax increased and Bcl-2 decreased in the model group,while the expression of Bax decreased and Bcl-2 increased after leonurine treatment.The results showed that leonurine inhibited the apoptosis of endometrial cells by regulating Bax and Bcl-2,and promoted the involution of uterus.
