摘 要:为探讨雷州黑鸭含溴结构域4蛋白(bromodomain containing 4,BRD4)基因的单核苷酸多态性与雷州黑鸭产蛋性能的关系,本试验以雷州黑鸭为研究对象,采用PCR扩增技术获得BRD4基因目的片段,通过直接测序检测BRD4基因单核苷酸多态位点(singlenucleotide polymorphisms, SNP),再使用HaploView 4.2软件对多个SNPs进行连锁不平衡分析获得单倍型,并分析BRD4基因的单倍型与雷州黑鸭产蛋性能的关联性。结果显示,在雷州黑鸭BRD4基因中发现4个突变位点,分别是I8-1897 T>A、I8-1936 A>C、I8-2046 C>T和I8-2109 T>C。其中,I8-1897 T>A位点的AA基因型个体的开产体质量显著低于TA基因型个体(P<0.05);I8-2046 C>T位点的CC基因型个体的开产体质量显著低于CT基因型个体(P<0.05);I8-2109 T>C的TT基因型个体在300天的产蛋数显著高与CC基因型个体(P<0.05);连锁分析发现,所有单倍型均与雷州黑鸭产蛋性能无显著差异(P>0.05)。结果表明,BRD4基因的I8-2109 T>C位点可作为雷州黑鸭繁殖性能的重要遗传标记。To investigate the relationship between the single nucleotide polymorphism of the Bromodomain containing 4(BRD4)gene of Leizhou Black Duck and the egg production performance of Leizhou Black Duck.Taking Leizhou Black Duck as the research object,PCR amplification technology was used to obtain the BRD4 gene target fragments,and the BRD4 gene single nucleotide polymorphisms(SNPs)were detected by direct sequencing,and then HaploView 4.2 software was used to analyze multiple SNPs.The haplotype was obtained by linkage disequilibrium analysis,and finally the correlation between the SNPs and haplotypes of the BRD4 gene and the laying performance of Leizhou black ducks was analyzed.The results of the study found that four mutation sites were found in the BRD4 gene of Leizhou black duck,namely I8-1897 T>A,I8-1936 A>C,I8-2046 C>T and I8-2109 T>C.I8-1897 T>A AA genotype individuals have significantly lower start weight than TA genotype individuals(P<0.05);I8-2046 C>T CC genotype individuals have significantly lower start weight than CT genotype individuals(P<0.05);I8-2109 T>C TT genotype individuals had significantly higher egg numbers at 300 days than CC genotype individuals(P<0.05);linkage analysis found that all haplotypes were similar to Leizhou Black Duck There was no significant difference in egg production performance(P>0.05).In conclusion,the I8-2109 T>C locus of BRD4gene can be used as an important genetic marker for the reproductive performance of Leizhou black duck.
