摘 要:目的:探讨大叶兰酚对人胃癌细胞株SGC-7901生长和侵袭的影响。方法:体外培养人胃癌细胞SGC-7901,加入不同浓度的大叶兰酚(0、30、60、120μmol/L),分别设为对照组与低、中、高剂量组,每组重复5孔。使用CCK-8检测法评估细胞活力,流式细胞术检测细胞凋亡情况,Transwell法检测细胞侵袭能力,并采用Westernblot检测各组热休克蛋白90(HSP90)、磷酸化蛋白激酶B(p-Akt)和细胞周期依赖性蛋白激酶1(CDK1)表达水平。结果:人胃癌细胞SGC-7901增殖率为高剂量组<中剂量组<低剂量组<对照组,差异有统计学意义(P<0.05)。人胃癌细胞SGC-7901凋亡率为高剂量组>中剂量组>低剂量组>对照组,差异有统计学意义(P<0.05)。侵入下室的人胃癌细胞数SGC-7901量为高剂量组<中剂量组<低剂量组<对照组,差异有统计学意义(P<0.05)。随着药物浓度的增加,中、高剂量组HSP90蛋白的表达水平,低、中、高剂量组p-Akt和CDK1蛋白表达下降,差异有统计学意义(P<0.05)。结论:大叶兰酚可抑制人胃癌细胞SGC-7901的生长、侵袭,并促进细胞凋亡,其作用可能与抑制HSP90/Akt/CDK1信号通路有关。Objective:To investigate the effect of gigantol on the growth and invasion of human gastric cancer cell line SGC-7901.Methods:Human gastric cancer cells SGC-7901 were cultured in vitro and added with gigantol at different concentrations(0,30,60,120μmol/L),as control group,low dose group,middle dose group and high dose group,with 5 duplicate wells per group.Cell viability was evaluated using CCK-8 assay.Cellular apoptosis was detected using flow cytometry.The invasion ability of cells was detected by Transwell method.The protein expression levels of heat shock protein 90(HSP90),phosphorylated protein kinase B(p-Akt),and cyclin dependent kinase 1(CDK1)in each group were detected by Western blot.Results:The proliferation rate of human gastric cancer cells SGC-7901 was ranked as high dose group<middle dose group<low dose group<control group,and the difference was statistically significant(P<0.05).The apoptosis rate of human gastric cancer cells SGC-7901 was ranked as high dose group>middle dose group>low dose group>control group,and the difference was statistically significant(P<0.05).The number of human gastric cancer cells SGC-7901 invading the lower chamber was ranked as high dose group<middle dose group<low dose group<control group,and the difference was statistically significant(P<0.05).With the increase of drug concentration,the HSP90 protein expression level in the middle dose group and high dose group decreased,while the p-Akt and CDK1 protein expression level in low dose group,middle dose group and high dose group decreased,and the difference was statistically significant(P<0.05).Conclusion:Gigantol may inhibit the growth and invasion of human gastric cancer cells SGC-7901 and promote apoptosis,and the mechanism is possibly related to the inhibition of HSP90/Akt/CDK1 signaling pathway.
