摘 要:为了分析新孢子虫在3种不同宿主细胞内的生长情况,该试验采用犬新孢子虫Nc-1株,将其与牛肾细胞(MDBK)、非洲绿猴肾细胞(Vero)和犬肾上皮细胞(MDCK)在37℃条件下培养10 d,每天观察比较3种细胞在虫体入侵前后的形态变化并记录虫体数量,绘制虫体生长曲线。结果表明:犬新孢子虫Nc-1株在MDBK、Vero和MDCK细胞中均能够正常生长。其中,Nc-1株在入侵Vero细胞的第6天可以较快达到生长高峰期,而在MDBK和MDCK细胞中生长较慢,在第7天才能达到虫体数量的最大值,且MDCK细胞中虫体数量最少。因此,除Vero细胞外,MDBK细胞也可作为一种培养新孢子虫良好的细胞系。该试验为深入了解新孢子虫感染机制和选择合适的体外培养模型提供重要参考。In order to analyze the growth of Neospora caninum in three different host cells,Nc-1 strain was used in this experiment and cultured with bovine kidney cells(MDBK),African green monkey kidney cells(Vero)and canine kidney epithelial cells(MDCK)at 37℃for 10 days.The morphological changes of three kinds of cells before and after invasion were observed and compared every day,and the number of invasive parasite was recorded,and the growth curve of invasive parasite was drawn.The results showed that Nc-1 strain could grow normally in MDBK,Vero and MDCK cells.Nc-1 strain could reach its growth peak faster on the 6th day after invasion of Vero cells,while it grew slowly in MDBK and MDCK cells,reaching the maximum number of invasive parasite on the 7th day,and the number of invasive parasite in MDCK cells was the least.Therefore,in addition to Vero cells,MDBK cells could also be used as a good cell line for culture N.caninum.This study provides an important reference for understanding the mechanism of N.caninum infection and selecting a suitable culture model in vitro.
