摘 要:为了探究Nesfatin-1对禁食小鼠多巴胺奖赏系统的影响,试验将40只C57BL/6小鼠随机分为阴性对照组、禁食24 h组、禁食48 h组、禁食24 h+Nesfatin-1组、禁食48 h+Nesfatin-1组,每组8只。禁食24 h、48 h组小鼠分别禁食24 h及48 h;禁食24 h+Nesfatin-1组及禁食48 h+Nesfatin-1组小鼠分别在禁食24 h及48 h基础上按体重120 mg/kg腹腔注射Nesfatin-1;阴性对照组小鼠腹腔注射等体积的PBS。注射Nesfatin-1后等待观察4 h,各组小鼠眼球采血后断颈处死,解剖取脑组织,采用ELISA方法检测各组小鼠血清Nesfatin-1及多巴胺含量;应用实时荧光定量PCR方法检测各组小鼠脑组织酪氨酸羟化酶(TH)基因和即刻早期基因(cFOS基因)的相对表达量。结果表明:与阴性对照组相比,禁食24 h组及禁食48 h组小鼠血清Nesfatin-1、多巴胺含量显著降低(P<0.05);与禁食24 h组相比,禁食24 h+Nesfatin-1组小鼠血清Nesfatin-1、多巴胺含量极显著升高(P<0.01);与禁食48 h组相比,禁食48 h+Nesfatin-1组小鼠血清Nesfatin-1、多巴胺含量极显著升高(P<0.01)。与阴性对照组相比,禁食24 h组及禁食48 h组小鼠脑组织TH基因的相对表达量极显著下降(P<0.01),cFOS基因的相对表达量极显著升高(P<0.01);与禁食24 h组相比,禁食24 h+Nesfatin-1组小鼠脑组织TH及cFOS基因相对表达量极显著升高(P<0.01);与禁食48 h组相比,禁食48 h+Nesfatin-1组小鼠脑组织TH及cFOS基因相对表达量显著升高(P<0.05)。说明Nesfatin-1对禁食小鼠多巴胺奖赏系统有一定影响。In order to explore the effects of Nesfatin-1 on the dopamine reward system in fasting mice,forty C57BL/6 mice were randomly divided into negative control group,fasting 24 h group,fasting 48 h group,fasting 24 h+Nesfatin-1 group and fasting 48 h+Nesfatin-1 group,with 8 mice in each group.Mice in the 24 h and 48 h fasting groups were fasted for 24 h and 48 h,respectively.Mice in fasting group for 24 h+Nesfatin-1 and fasting group for 48 h+Nesfatin-1 were intraperitoneally injected with Nesfatin-1 according to 120 mg/kg body weight on the basis of fasting for 24 h and 48 h,respectively.Mice in negative control group were intraperitoneally injected with equal volume PBS.After injection of Nesfatin-1 and waiting for observation for 4 h,the mice in each group were sacrificed after eyeball blood collection,and then the brain tissue was dissected.The contents of Nesfatin-1 and dopamine in serum of mice in each group were detected by ELISA.The relative expression levels of tyrosine hydroxylase(TH)and immediate early gene(cFOS)in brain tissues of mice in each group were detected by real-time fluorescence quantitative PCR.The results showed that compared with the negative control group,the contents of Nesfatin-1 and dopamine in the serum of mice in the fasting 24 h group and the fasting 48 h group were significantly decreased(P<0.05).Compared with the fasting 24 h group,the serum levels of Nesfatin-1 and dopamine in the fasting 24 h+Nesfatin-1 group were extremely significantly increased(P<0.01).Compared with the fasting 48 h group,the serum levels of Nesfatin-1 and dopamine in the fasting 48 h+Nesfatin-1 group were extremely significantly increased(P<0.01).Compared with the negative control group,the relative expression of TH mRNA in the brain tissue of mice in the fasting 24 h group and the fasting 48 h group decreased extremely significantly(P<0.01),and the relative expression of cFOS mRNA increased extremely significantly(P<0.01).Compared with fasting 24 h group,the relative expression of TH and cFOS mRNA in brain tiss
