摘  要:为了探究TYR基因对中国家驴毛色的影响,试验利用PCR-SSCP方法对14个品种408个毛色表型记录清晰的中国家驴TYR基因第一外显子的多态性与毛色的关系进行了研究,并根据其毛色进行分类,计算不同毛色的基因型频率和基因频率。结果表明:中国家驴TYR基因在第一外显子存在g.259A→G突变位点,由此产生了三种基因型AA、AG和GG,该位点为异亮氨酸到缬氨酸的非同义突变。AA基因型频率在乌头驴、黑三粉驴、青三粉驴和灰三粉驴中的分布逐渐降低,GG基因型频率分布逐渐提高,2头白驴均为GG基因型;等位基因A和G的频率在灰三粉驴中相等,在其他毛色中,等位基因A的频率大于G。说明随着中国家驴被毛中黑色被毛分布的减少,AA基因型频率逐渐下降,GG基因型频率逐渐提高,该位点与中国家驴黑白被毛的形成有关。In order to explore the effect of TYR gene on the hair color of Chinese donkeys,in this experiment,the relationship between the polymorphisms of the first exon of the TYR gene of Chinese donkey TYR gene and hair color in 408 hair color phenotypes of 14 breeds was studied by PCR-SSCP method,and classified according to their different hair colors.The genotype frequency and gene frequency of different hair colors were calculated.The results showed that the Chinese donkey TYR gene had a g.259A→G mutation site in the first exon,resulting in three genotypes:AA,AG and GG,which were nonsynonymous mutations from isoleucine to valine.The frequency distribution of AA genotype in black-head,black-pink,green-pink and gray-pink donkey decreased successively,and the frequency distribution of GG genotype gradually increased.Both white donkeys had a genotype of GG.The frequencies of alleles A and G were equal in the gray-pink donkey,and in other hair colors,the frequency of allele A was greater than that of G.The results suggested that with the decrease of the distribution of black hair in donkey hair in China,the frequency of AA genotype gradually decreased,and the frequency of GG genotype gradually increased,which was related to the formation of black and white hair in Chinese donkeys.
