摘  要:红螯螯虾虹彩病毒(Cherax quadricarinatus iridovirus,CQIV)是以甲壳动物为主要宿主的线性双链DNA病毒。CQIV对宿主的致死率极高,且传播到我国多个省份养殖场,已成为虾类养殖的重大威胁。然而,目前对该病毒结构蛋白尤其是囊膜蛋白的组成还不清楚。本研究利用液相色谱偶联质谱技术,从纯化的CQIV病毒粒子中鉴定到60个病毒结构蛋白,并选取168L作进一步分析。生物信息学分析表明168L含有3个跨膜结构域。表达时相和药物抑制分析表明168L为病毒晚期基因。免疫印记分析表明168L存在于完整病毒粒子和病毒膜组分中。免疫电镜结果表明168L为病毒囊膜蛋白。本研究结果将为阐明CQIV的结构和感染机制提供新的线索。Cherax quadricarinatus iridovirus(CQIV)is a crustacean virus with icosahedral virion that contains linear double-stranded DNA.CQIV has been a potential threat to shrimp industries which has spread to several provinces of China and infects with a high mortality.However,the viral structural proteins especially the envelope proteins of CQIV remain largely uncharacterized.In this study,the intact virion was directly used for shotgun-proteome analysis to further characterize the protein components of the virus.This led to identification of 60 viral proteins,of which,168L was chosen for further analysis.Temporal analysis and drug inhibition assay revealed that 168L is a late expression gene.The specific antiserum against 168L was raised and Western Blot showed that it bound specifically to a 57 kD protein in both the intact virions and the envelope fraction.Immunogold labeling revealed that 168L is a component of the viral envelope.in this investigation lead to a better understanding of the CQIV structural proteins.
