电针联合泮托拉唑对乳腺癌裸鼠免疫功能、 病理形态及CYP17/CYP19表达的影响

摘　　要：目的:探讨电针联合泮托拉唑对乳腺癌裸鼠免疫功能、病理形态及CYP17/CYP19表达的影响。方法:45只裸鼠分为对照组(n=9)、乳腺癌组(n=8)、药物组(n=9)、穴位治疗组(n=8)及联合组(n=8),除对照组外均建立乳腺癌动物模型,建模成功后,药物组及联合组灌胃泮托拉唑,穴位治疗组及联合组进行“百会”“长强”穴位针刺治疗,观察各组裸鼠血清中雌激素及血清NK细胞及T淋巴细胞受体水平;计算各组裸鼠瘤体质量;HE染色观察肿瘤组织病理形态;TUNEL法检测各组裸鼠肿瘤组织细胞凋亡率;免疫印记检测肿瘤组织CYP17、CYP19、Bcl-2及Bax蛋白。结果:与对照组相比,乳腺癌组裸鼠血清E2、P、PR、LH、FSH、CD3^(+)、FOXP3^(+)、CYP17、CYP19及Bcl-2水平升高,TCRαβ^(+)、CD161a^(+)抗体、T淋巴细胞CD3^(+)CD4^(+)、CD3^(+)CD8^(+)及Bax降低,差异具有统计学意义(P<0.05);与乳腺癌组相比,药物组及穴位治疗组血清E2、P、PR、LH、FSH、CD3^(+)、FOXP3^(+)、CYP17、CYP19及Bcl-2水平降低,TCRαβ^(+)、CD161a^(+)抗体、T淋巴细胞CD3^(+)CD4^(+)、CD3^(+)CD8^(+)及Bax升高,差异具有统计学意义(P<0.05),与穴位治疗组和联合组相比存在差异,差异具有统计学意义(P<0.05);对照组、乳腺癌组、药物组、穴位治疗组及联合组的肿瘤质量分别为(0.00±0.00)g、(0.42±0.06)g、(0.30±0.05)g、(0.28±0.06)g及(0.21±0.03)g,多组间比较差异有统计学意义(F=78.340,P<0.05);对照组、乳腺癌组、药物组、穴位治疗组及联合组的肿瘤组织细胞凋亡率分别为(0.00±0.00)%、(3.51±0.71)%、(24.12±5.22)%、(24.28±6.22)%及(45.78±8.44)%,多组间比较,差异具有统计学意义(F=106.100,P<0.05);乳腺癌组裸鼠乳腺腺体结构消失,小叶增加且导管出现扩展,腺体出现瘤样改变;与乳腺癌组相比,药物组及穴位治疗组乳腺小叶结构改善,导管增生好转;联合组乳腺导管增生及扩张明显改善,小叶腺泡减小,瘤样变化程度减弱。�Objective:To investigate the effects of electro-acupuncture(EA)combined with Pantoprazole on immune function,pathological morphology and expression of CYP17/CYP19 in nude mice with breast cancer.Methods:45 nude mice were divided into the control group(n=9),the model group(n=8),the medication group(n=9),the acupoints treatment group(n=8)and the combination group(n=8).After successful modeling of breast cancer,the medication group was given Pantoprazole gavage,the acupoints treatment group was treated with electro-needling Baihui(DU20)and Changqiang(DU1),and the combination group was treated with Pantoprazole gavage and electro-needling DU20 and DU1.The serum levels of estrogen,NK cells and T lymphocyte receptors were observed.The tumor mass of nude mice was calculated.HE staining was used to observe the pathological morphology of tumor tissues.TUNEL method was used to detect the apoptosis rate of tumor cells in each group.Immunoblotting method was performed to detect the proteins of CYP17,CYP19,Bcl-2 and Bax in tumor tissues.Results:Compared with those in the control group,the serum levels of E2,P,PR,LH,FSH,CD3^(+),FOXP3^(+),CYP17,CYP19,Bcl-2 were increased,TCRαβ^(+),CD161a^(+)antibody,T lymphocyte CD3^(+)CD4^(+),CD3^(+)CD8^(+)and Bax were decreased in the model group(P<0.05).Compared with those in the model group,the serum levels of E2,P,PR,LH,FSH,CD3^(+),FOXP3^(+),CYP17,CYP19 and Bcl-2 were decreased,and TCRαβ^(+),CD161a^(+)antibody,T lymphocytes CD3^(+)CD4^(+),CD3^(+)CD8^(+)and Bax were increased in the medication group and the acupoints treatment group(P<0.05);in which there were statistical differences between the acupoints treatment group and the combination group(P<0.05).The tumor mass was(0.00±0.00)g,(0.42±0.06)g,(0.30±0.05)g,(0.28±0.06)g and(0.21±0.03)g in the control group,the model group,the medication group,the acupoints treatment group and the combination group,respectively;in which there were statistical differences among the groups(F=78.340,P<0.05).The apoptosis rate of tumor cells was(0.00
