摘  要:建立了永停滴定法测定过氧化氢酶活性的新方法。在盐酸介质中,硫酸铈铵(Ce^(4+))与H_(2)O_(2)发生2∶1的定量反应,当用Ce^(4+)标准溶液滴定H_(2)O_(2)时,酶分解前后H_(2)O_(2)减少量决定过氧化氢酶活性。结果表明,过氧化氢酶活量X在0.000~141 U范围内与Ce^(4+)标准溶液滴定体积差呈线性关系:ΔV(Ce^(4+))=0.2010X-0.0679,相关系数为0.9994,方法的最低检出量为0.400 U。方法用于草鱼活体组织中过氧化氢酶活性测定。In this paper,a new method for the determination of catalase activity by dead stop titration was established.In hydrochloric acid medium,the quantitative reaction of ceric ammonium sulfate(Ce^(4+))with H_(2)O_(2) is 2∶1.When H_(2)O_(2) was titrated with Ce^(4+)standard solution,the activity of catalase was determined by the change of H_(2)O_(2) before and after enzyme decomposition.The results showed that there was a linear relationship between enzyme activity X and Ce^(4+)standard solution,and the titration volume difference in the range of 0.000~141UΔV(Ce^(4+))=0.2010X-0.0679.Meanwhile,the correlation coefficient was 0.9994 and the check out the quantity was 0.400 U.The method was applied to the determination of catalase activity in living tissues of grass carp.
