摘  要:铜绿假单胞菌的耐药形势日益严峻,通过耐药泵将药物排出是铜绿假单胞菌产生耐药性的重要原因之一。采用生物信息学方法对PA3573基因编码蛋白进行结构功能预测以及编码蛋白与鲎素肽分子对接的模拟实验预测,并对PA3573基因在铜绿假单胞菌抗鲎素突变株中转录表达情况进行分析。结果表明,PA3573基因编码蛋白是碱性带正电的稳定非分泌的疏水性跨膜蛋白,主要含有α-螺旋结构、无信号肽和21个潜在的磷酸化位点。PA3573蛋白含有一个主要协同转运超家族(MFS)保守结构域,属于主要协同转运超家族。该蛋白与DR97-4591蛋白基因共表达,与阴沟肠杆菌、大肠杆菌等有较近的亲缘关系。研究发现PA3573基因在不同传代浓度诱导下获得的抗鲎素突变株中均有明显差异表达,且预测该编码蛋白与鲎素通过氢键和疏水作用相结合的方式发挥作用。该结果为进一步阐明PA3573基因编码蛋白在铜绿假单胞菌中的作用提供一定的理论指导。The drug resistance situation of Pseudomonas aeruginosa is becoming increasingly severe.Drug excretion through drug resistance pumps is one of the important reasons for the development of drug resistance in Pseudomonas aeruginosa.Bioinformatics methods were used to predict the structure and function of the protein encoded by the PA3573 gene and simulated experimental predictions of molecular docking between the encoded protein and limulus peptide,and the transcriptional expression of the PA3573 gene in the limululin-resistant mutant strain of Pseudomonas aeruginosa was analyzed.The results showed that the protein encoded by PA3573 gene was a stable non-secreted hydrophobic transmembrane protein with positive basic charge,which mainly contains a helical structure,no signal peptide and 21 potential phosphorylation sites.The PA3573 protein contains a major facilitator superfamily(MFS)conserved domain and belongs to the major facilitator superfamily.This protein is co-expressed with the DR97-4591 protein gene and is closely related to Enterobacter cloacae and Escherichia coli.The study found that the PA3573 gene was significantly differentially expressed in the limulus-resistant mutant strains induced by different passage concentrations,and it was predicted that the encoded protein interacts with limulus through hydrogen bonds and hydrophobic interactions.This result provides certain theoretical guidance for further elucidating the role of the protein encoded by the PA3573 gene in Pseudomonas aeruginosa.
