清营颗粒对大鼠的急性毒性研究

摘　　要：为探究清营颗粒的急性毒性效应,选用SD大鼠为研究对象,采用限量法进行试验,经口单次给予雌雄各10只SD大鼠受试品清营颗粒,给药剂量为30 g/kg体重,然后分别于染毒前0 d、染毒后1、2、7、14 d测定大鼠体重,并连续14 d进行详细临床观察。观察期结束后,对大鼠进行大体剖检。雌雄大鼠在给予30 g/kg体重剂量的清营颗粒后,精神状态良好,采食、大小便及体重增长正常,直至试验结束时无异常临床症状出现。雄性大鼠在给药1、2、7、14d后,体重持续增长;仅有4只雌性大鼠在给药1 d或2 d后体重降低,其他雌性大鼠体重持续增长。剖检结果显示,无肉眼可见病变。综上所述,清营颗粒对雌性和雄性SD大鼠的经口急性毒性耐受剂量大于30 g/kg体重。根据化学物急性毒性(LD_(50))剂量分级标准依据,表明清营颗粒无毒。To study the acute toxicity of Qingying Granules in rats and provide evidence for the research and development of new drugs.SD rats were selected as the study subjects and tested using a limited-dose method.Ten male and ten female SD rats were given a single oral dose of the test article Qingying Granules at a dose of 30 g/kg body weight.Then,weight measurements were conducted at D0 before exposure,and at D1,D2,D7,and D14 after exposure.Detailed clinical observations were performed for 14 consecutive days.After the observation period,the experimental animals were subjected to gross dissection.After receiving a 30 g/kg body weight dose of Qingying Granules,male and female rats showed good mental status,normal diet,normal urination and defecation,and normal weight gain.Until the experiment ended 14 days after administration,no clinical abnormalities were observed.Male animals continued to gain weight on days 1,2,7,and 14 after administration.Four female animals experienced weight loss on day 1 or 2 after administration,while the rest continued to gain weight.The animal autopsy showed no visible lesions.The oral acute toxicity tolerance dose of Qingying Granules in female and male SD rats is greater than 30g/kg body weight.According to the acute toxicity(LD_(50))dose classification of chemicals,Qingying Granules is non-toxic.
