摘 要:目的优化炒水红花子的工业化生产参数,考察炒水红花子的质量。方法采用正交分析与工业化生产相结合,采用独立性权重法进行综合评分,对炒水红花子工业化生产参数进行优化。以最优炮制工艺,炮制10批不同产地的水红花子,采用高效液相色谱法(HPLC)测定炮制品中花旗松素的含量,薄层色谱法对炮制品进行考察。结果炒水红花子的最优生产工艺为置270℃预热炒锅内,炒制5 min,转速15 r/min。HPLC法测定炒水红花子中花旗松素的含量,进样量在0.04~2.00μg范围内,线性关系良好(r=0.9999),平均回收率为100.3%(RSD=2.3%,n=9),含量结果在0.13%~0.31%之间。薄层色谱法能进行有效鉴别。结论本研究明确了炒水红花子最优工业化生产参数,考察10批不同产地炮制品质量,验证生产工艺稳定可靠。对炒水红花子质量监管和地方标准升级为国家标准提供参考。Objective To optimize the industrial production parameters and investigate the quality of Polygoni Orientalis Fructus Tostum.Methods The orthogonal analysis was combined with industrial production,and the independent weight method was used for comprehensive scoring to optimize the industrial production parameters of Polygoni Orientalis Fructus Tostum.According to the optimal processing process,10 batches of Polygoni Orientalis Fructus Tostum from different re-gions were processed.The content of taxifolin in the processed products was determined by high performance liquid chro-matography(HPLC),and the processed products were investigated by thin layer chromatography.Results The optimal pro-duction process for Polygoni Orientalis Fructus Tostum was to place them in a pre-heated wok at 270℃for 5 min at a speed of 15 r/min.HPLC method was used to determine the content of taxifolin in Polygoni Orientalis Fructus Tostum.The injection volume was in the range of 0.04-2.00μg,and the linear relationship was good(r=0.9999).The average recovery rate was 100.3%(RSD=2.3%,n=9),and the content results were in the range of 0.13%-0.31%.Thin layer chromatography can be used for effective identification.Conclusion In this study,the optimal industrial production parameters of Polygoni Orientalis Fructus Tostum were determined,and the quality of 10 batches of processed products from different origins was investigated to verify that the production process was stable and reliable.It provides reference for the quality supervision of Polygoni Orientalis Fructus Tostum and upgrading of local standards to national standards.
