摘  要:目的高效液相色谱法拆分顺式双环[3.2.0]庚-2-烯-6-酮对映异构体并进行含量测定,同时进行含量测定的方法学研究。方法采用硅胶表面涂覆有直链淀粉-三(S)-α-甲基苯基氨基甲酸酯手性固定相,以正己烷-异丙醇(体积比95∶5)为流动相,流速0.8 mL·min^(-1),柱温25℃,检测波长210 nm,采用外标法测定顺式双环[3.2.0]庚-2-烯-6-酮对映异构体的含量,采用面积归一化法计算对映异构体超量值。结果顺式双环[3.2.0]庚-2-烯-6-酮对映异构体分离度为11.4,两个对映异构体在质量浓度2~40 mg·L^(-1)内有良好线性关系(R^(2)≥0.999),(+)-顺式双环[3.2.0]庚-2-烯-6-酮低、中、高质量浓度平均加样回收率(RSD)分别为99.16%(0.8%)、99.34%(0.8%)、100.2%(0.7%);(-)-顺式双环[3.2.0]庚-2-烯-6-酮低、中、高质量浓度平均加样回收率RSD分别为99.72%(0.9%)、100.0%(0.3%)和98.81%(0.6%)。结论该方法能够用面积归一化法准确测定顺式双环[3.2.0]庚-2-烯-6-酮对映异构体超量值,为前列腺素类药物手性中间体提供新的分离方法。Objective To enantioseperate bicyclo[3.2.0]hept-2-en-6-oneenantiomers and determine their contants by HPLC,and to study the HPLC method of content determination simultaneously.Methods The separation was developed on the chiral stationary phase silicon which the surface is coated with amylose-three(S)-α-methyl phenyl carbamate.The mobile phase was n-hexane-isopropyl alcohol(95∶5,V/V)at the flow rate of 0.8 mL·min^(-1).The column temperature was set at 25℃,and the detection wavelength was set at 210 nm.The contents of bicyclo[3.2.0]hept-2-en-6-oneenantiomers were determined by external standard method and the enantiomeric excess was determined by area normalization method.Results The resolution of the two enantiomers was 11.4.The linear ranges of the two enantiomers were 2~40μg·ml^(-1)(R^(2)≥0.999).The average recovery rates(RSDs)of(+)-bicyclo[3.2.0]hept-2-en-6-one at low,medium and high concentration were 99.16%(0.8%),99.34%(0.8%),100.2%(0.7%).The average recovery rates(RSDs)of(-)-bicyclo[3.2.0]hept-2-en-6-one at low,medium and high concentration were 99.72%(0.9%),100.0%(0.3%),98.81%(0.6%).Conclusion The HPLC method has the advantages of simple operation,good resolution,and high accuracy,and can determine the enantiomers excess of bicyclo[3.2.0]hept-2-en-6-one by area normalization method simultaneously.It provides a new method for the separation of prostaglandins chiral intermediates.
