摘  要:为了解决两步法制备常规温敏型复合水凝胶时胶凝速率难以控制、表面凝胶快且致密、交联结构不均匀影响水凝胶溶胀性能和力学性能等问题,选用N-异丙基丙烯酰胺单体(NIPAm)和海藻酸钠(Alg)为主要材料,使用CaCO_(3)和D-葡萄糖酸内酯(GDL)生成的CaCO_(3)-GDL体系作为钙源为Alg提供离子交联剂,通过一步法制备海藻酸钙-聚(N-异丙基丙烯酰胺)(PNIPAm)交联互穿网络结构温敏型水凝胶(PAGel)。通过傅里叶红外光谱仪(FTIR)、扫描电镜(SEM)、流变仪和差示扫描量热仪(DSC)对水凝胶的结构、形貌和体积相转变温度(VPTT)进行表征,探讨不同NIPAm单体和交联剂N,N-亚甲基双丙烯酰胺(MBAA)含量对水凝胶溶胀性能和温敏收缩性能的影响。结果表明:综合各种性能筛选出的PAGel的VPTT均在32℃附近,CaCO_(3)-GDL缓释体系下的水凝胶结构呈现网络均匀而致密的蜂窝状结构,力学性能优良,其溶胀质量比、VPTT和温敏收缩率分别为29.1、33.1℃和35.37%,该结构特性可减少化学药物的负载与释放,同时利用体积收缩的物理方法促进创面周围的皮肤组织收缩,为创造低毒性愈合微环境和加快伤口愈合提供可能性。In order to solve the problems such as difficult to control the gelation rate,fast and dense surface gel and uneven cross-linking structure affecting the swelling and mechanical properties of hydrogels during the preparation of conventional temperature-sensitive composite hydrogels by two-step method,N-isopropylacrylamide monomer(NIPAm)and sodium alginate(Alg)were selected as the main materials,and CaCO_(3)-GDL system produced by CaSO 4 and D-gluconolactone(GDL)was used as calcium source to provide ionic crosslinking agent for Alg.Temperature sensitive hydrogel(PAGel)with calcium alginate poly(n-isopropylacrylamide)(PNIPAm)cross-linked interpenetrating network structure was prepared by one-step method.The structure,morphology and volumetric phase transition temperature(VPTT)of hydrogels were characterized by Fourier infrared spectroscopy(FTIR),scanning electron microscopy(SEM),rheometer and differential scanning calorimeter(DSC).The effects of different NIPAm monomer and cross-linker N,N-methylene bisacrylamide(MBAA)contents on swelling and temperature-sensitive shrinkage properties were investigated.The results show that:The VPTT of PAGel selected by various properties is all near 32℃.The hydrogel structure of CaCO_(3)-GDL slow release system presents a uniform network and dense honeycomb structure with excellent mechanical properties.The swelling mass ratio,VPTT and temperature sensitive shrinkage rate are 29.1,33.1℃and 35.37%,respectively.This structural property can reduce the load and release of chemical drugs,and promote the contraction of skin tissue around the wound by physical methods of volume contraction,providing the possibility of creating a low-toxic healing microenvironment and accelerating wound healing.
