摘  要:目的本研究旨在探究0.7μg/kg短链氯化石蜡(SCCPs)暴露对昆明小鼠肝脏功能及其代谢的影响。方法雄性昆明小鼠随机分为对照组、SCCPs暴露组,每组各8只,48d后收集肝脏,检测肝功能生化指标并进行组织病理学分析、Hoechst 33258荧光观察和能量代谢相关检测。结果与对照组比较,SCCPs暴露组肝功能生化指标显著升高(P<0.05),肝细胞肿大,可见空泡化病理结构,肝细胞线粒体形态和结构损伤,线粒体氧化代谢紊乱。结论长期低剂量SCCPs暴露可引起小鼠肝功能及代谢异常,作用机制或与其致肝细胞线粒体功能障碍有关。Objective The aim of this study was to investigate the effects of 0.7μg/kg short-chain chlorinated paraffins(SCCPs)exposure on liver function and its metabolism in Kunming mice.Methods Male Kunming mice were randomly divided into control and SCCPs-exposed groups(n=8 mice),and livers were collected after 48 days for the biochemical indices of liver function and histopathological analysis,Hoechst 33258 fluorescence observation and energy metabolism-related assays.Results Compared with the control group,the SCCPs-exposed group showed significantly higher biochemical parameters of liver function(P<0.05),hepatocyte enlargement,visible vacuolated pathological structures,damaged hepatocyte mitochondrial morphology and structure,and disrupted mitochondrial oxidative metabolism.Conclusion Long-term low-dose SCCPs exposure could still cause abnormal liver function and metabolism in mice,and the mechanism may be related to mitochondrial dysfunction in hepatocytes caused by SCCPs exposure.
