摘  要:目的:探讨参芪蛭龙汤对膜性肾病小鼠肾小球足细胞凋亡的影响。方法:制备阳离子化牛血清白蛋白(C-BSA),复制膜性肾病小鼠模型。将40只模型小鼠随机分为模型组、参芪蛭龙汤高、低剂量组和雷公藤多苷片组,另取10只正常小鼠作为正常组。连续给药4周后,处死小鼠,HE、Masson、PAS染色观察肾组织病理变化;RT-PCR检测肾组织Bcl-2(B淋巴细胞瘤-2)、Bax(/BCL2-相关X蛋白)、Caspase-3(半胱氨酸天冬氨酸特异性蛋白酶3)mRNA表达变化;Western blot检测肾组织Bcl-2、Bax、Caspase-3蛋白表达变化。结果:HE染色发现,与模型组比较,参芪蛭龙汤高剂量组肾小球体积减小,仅有炎性细胞浸润减少;Masson染色可见,与模型组比较,各给药组可见肾小球体积减小,基底膜增厚及组织纤维化减轻;PAS染色可见,各给药组基底膜病变较模型组显著减轻,参芪蛭龙汤高剂量组并未出现明显基底膜增厚等改变。RT-PCR结果显示,与模型组比较,参芪蛭龙汤各组小鼠肾组织Bax、Caspase-3 mRNA表达减少(P<0.01),Bcl-2 mRNA表达增加(P<0.01);Western blot结果发现,与模型组比较,参芪蛭龙汤高剂量组小鼠肾组织Bax、Caspase-3蛋白表达减少(P<0.01),Bcl-2蛋白表达增加(P<0.01)。结论:参芪蛭龙汤能够改善膜性肾病小鼠肾组织病变,抑制肾小球足细胞凋亡。Objective:To investigate the effects of Shenqi Zhilong Decoction on the apoptosis of glomerular podocytes in mice with membranous nephropathy.Methods:Cationized bovine serum albumin(C-BSA)was prepared to replicate the mouse model of membranous nephropathy.40 model mice were randomly divided into model group,Shenqi Zhilong Decoction high dose group,low dose group and Tripterygium wilfordii polyglycoside tablet group,and 10 normal mice were selected as normal group.After 4 weeks of continuous administration,the mice were killed,and the pathological changes of renal tissue were observed by HE,Masson,PAS staining.The mRNA expressions of Bcl-2,Bax,Caspase-3 were detected by RT-PCR.Western blot analysis was used to determine renal Bcl-2,Bax,Caspase-3 protein expressions.Results:HE staining showed that compared with model group,the glomerular volume of Shenqi Zhilong Decoction high-dose group was decreased,only the inflammatory cell infiltration was decreased;Masson staining showed that compared with model group,the volume of glomerulus was decreased,basement membrane got thickened and tissue fibrosis wasdecreased in each administration group.PAS staining showed that the pathological changes of basement membrane in each administration group was significantly reduced compared with that in model group,but the thickening of basement membrane did not appear in Shenqi Zhilong Decoction high-dose group.RT-PCR results showed that compared with model group,the mRNA expressions of Bax and Caspase-3 in kidney tissues of Shenqi Zhilong Decoction group were decreased(P<0.01),while the mRNA expression of Bcl-2 was increased(P<0.01).Western blot results showed that compared with model group,the expressions of Bax and Caspase-3 protein in kidney tissue of Shenqi Zhilong Decoction high-dose group were decreased(P<0.01),while the expression of Bcl-2 protein was increased(P<0.01).Conclusion:Shenqi Zhilong Decoction can improve the renal tissue lesion and inhibit the apoptosis of glomerular podocytes in mice with membranous nephropathy.
