摘 要:目的研究RANKL/RANK/OPG信号通路在小鼠膝关节假体无菌性松动中的作用机制。方法28只小鼠以计算机随机数字表法分成正常组、实验组,最终均分别纳入12只。实验组通过在胫骨近端骨髓腔中注射钴铬颗粒悬液建立小鼠膝关节假体无菌性松动模型,正常组注射等量等渗盐水。采用实时荧光定量PCR(qRT-PCR)法检测界膜组织中RANKL、RNAK、OPG mRNA表达量;免疫组化染色法检测界膜组织中RANKL、RNAK、OPG、抗酒石酸酸性磷酸酶(TRAP)蛋白表达量。结果与正常组比较,实验组建模后6、12周界膜组织RANKL、RNAK mRNA及蛋白表达量,RANKL/OPG升高,OPG mRNA及蛋白表达量降低(P<0.05)。与建模后6周比较,正常组建模后12周界膜组织RANKL、RNAK mRNA及蛋白表达量及RANKL/OPG降低,OPG mRNA及蛋白表达量升高(P<0.05),实验组界膜组织RANKL、RNAK mRNA及蛋白表达量及RANKL/OPG升高,OPG mRNA及蛋白表达量降低(P<0.05)。与正常组比较,实验组建模后6、12周界膜组织TRAP蛋白表达量升高(P<0.05)。与建模后6周比较,正常组建模后12周界膜组织TRAP蛋白表达量降低(P<0.05),实验组界膜组织TRAP蛋白表达量升高(P<0.05)。结论小鼠膝关节假体无菌性松动的界膜组织中RANKL、RNAK表达量及RANKL/OPG升高,OPG表达量降低,RANKL/RNAK/OPG信号通路表达失衡与其发生、发展相关。Objective To study the mechanism of RANKL/RANK/OPG signaling pathway in the aseptic loosening of knee prosthesis in mice.Methods 28 mice were divided into normal group and experimental group by computer random number table,and 12 mice were included in each group at last.A mouse model of aseptic loosening of knee prosthesis was established by injecting cobalt chromium granule suspension into the bone marrow cavity of the proximal tibia,while the normal group was injected with the same amount of isotonic saline.The mRNA expressions of RANKL/RNAK/OPG in peripheral membrane tissues were detected by real-time quantitative fluorescent PCR(qRT-PCR);the expression levels of RANKL/RNAK/OPG and tartrate-resistant acid phosphatase(TRAP)were detected by immunohistochemical staining.Results Compared with the normal group,the mRNA and protein expression levels of RANKL and RNAK in the peripheral membrane tissues,and RANKL/OPG of the experimental group were increased at the 6th and 12th week after modeling,while the mRNA and protein expression levels of OPG were decreased(P<0.05).Compared with the 6th weeks after modeling,the mRNA and protein expression levels of RANKL and RNAK in the peripheral membrane tissues at the 12th week after modeling in the normal group were decrease,while the mRNA and protein expression levels of OPG were increased(P<0.05),while the experimental group showed opposite results.Compared with the normal group,the expression of TRAP protein in the peripheral membrane tissues of the experimental group increased at 6th and 12th week after modeling(P<0.05).Compared with the result of the 6th weeks after modeling,the expression of TRAP protein in peripheral membrane tissues at the 12th week after modeling in the normal group decreased(P<0.05),but the expression increased in the experimental group(P<0.05).Conclusion The expression levels of RANKL,RNAK and RANKL/OPG were increased and the expression levels of OPG were decreased in the aseptic loosening of the knee prostheses in mice.The imbalance of RANKL/RNAK/
