摘 要:以水生黄杆菌作为出发菌株,研究了烷基葡萄糖胺对水生黄杆菌的细胞活性、形态、细胞膜通透性和胞外分泌维生素K_(2)的影响。结果表明,当在培养基中添加0.6%的烷基葡萄糖胺可提高维生素K_(2)的合成浓度。发酵到第8 d时,水生黄杆菌发酵合成胞外维生素K_(2)浓度达到18.7 mg/L,此时测得发酵合成维生素K_(2)总产量为25.3 mg/L,较未添加烷基葡萄糖胺时提高了204.82%。发酵液表面张力由37.4 mN/m降低到36.6 mN/m,利用扫描电镜观察发现烷基葡萄糖胺改变了菌体的细胞形态,流式细胞术分析发现菌株的细胞膜通透性增加。说明烷基葡萄糖胺可促进维生素K_(2)的胞外分泌,为微生物发酵法促进维生素K_(2)跨膜转运提供了参考。The effects of alkyl glucosamine on the cell viability,morphology,cell membrane permeability and extracellular secretion of vitamin K_(2) by taking Flavobacterium hydatis as the starting strain.The results showed that the addition of 0.6%alkyl glucosamine in the medium could enhance the concentration of vitamin K_(2).The concentration of extracellular vitamin K_(2) was 18.7 mg/L,the concentration of total vitamin K_(2) was increased to 25.3 mg/L,which was 204.82%higher than untreated control after fermentation on the 8th day.The surface tension of the fermentation broth decreased from 37.4 mN/m to 36.6 mN/m.The result of scanning electron microscopy showed that alkyl glucosamine changed the cell morphology of the bacteria.Flow cytometry analysis showed that the cell membrane permeability was increased.It explained that alkyl glucosamine could promote the exocytosis of vitamin K_(2),also provided a reference for promoting the extracellular secretion of vitamin K_(2) by microbial fermentation.
