蒙药清心沉香八味散对急性抑郁模型小鼠行为学及单胺类神经递质的影响

摘　　要：目的:观察蒙药清心沉香八味散的抗抑郁作用及对单胺类神经递质的影响。方法:将实验动物(雄性昆明种小鼠)随机分为5组,分别为空白对照组(蒸馏水)、阳性药对照组(盐酸氟西汀0.02 g/kg)及受试蒙药清心沉香八味散高、中、低剂量组(27、9、3 g/kg),各组小鼠采用灌胃方式给药,7 d后采用小鼠悬尾实验(TST)、小鼠强迫游泳实验(FST)初步评估其抗抑郁作用。FST行为学实验完成后取血浆及海马组织,采用酶联免疫吸附方法(ELISA)分别检测动物血浆及海马组织中5-HT、DA、NE的含量。结果:与空白组比较,蒙药中、低剂量组能显著缩短小鼠FST的行为绝望时间,差异具有统计学意义(P<0.05),蒙药低剂量组能显著缩短小鼠TST的行为绝望时间,差异具有统计学意义(P<0.05),蒙药清心沉香八味散低剂量组能显著升高小鼠血浆中DA水平,差异具有统计学意义(P<0.05),蒙药清心沉香八味散低、中、高剂量组能显著升高小鼠海马组织中5-HT、DA、NE的含量,差异具有统计学意义(P<0.05)。结论:蒙药清心沉香八味散对小鼠可能有一定的抗抑郁样作用,其作用可能与其调节单胺类神经递质的含量相关。Objective:To investigate the antidepressive effect of Qingxin Chenxiang Bawei Powder and its effect on monoamine neurotransmitters.Methods:Kunming male mice were randomly divided into the blank control group(distilled water),positive-drug group(fluoxetine hydrochloride 0.02 g/kg)and Mongolian medicine Qingxin Chenxiang Bawei Powder groups.Intragastric administration were performed on all experimental mice.Mice tail suspension test(TST)and forced swimming test(FST)were used to evaluate the antidepressive effect of the drug 7 days after intragastric administration.After FST behavioral experiment,the contents of 5-HT,DA and NE in plasma and hippocampus were determined by enzyme-linked immunosorbent assay(ELISA).Results:Compared with the blank control group,medium-dose and low-dose groups of Mongolian medicine could significantly shorten the immobility time in mice FST,and the difference was statistically significant(P<0.05).The immobility time in mice TST was significantly shortened in the low-dose group,and the difference was statistically significant(P<0.05).The plasma DA level was significantly increased in the low-dose group,and the difference was statistically significant(P<0.05).The levels of 5-HT,DA and NE in the hippocampus of mice in the low-dose,medium-dose and high-dose groups were significantly increased,and the difference was statistically significant(P<0.05).Conclusion:Qingxin Chenxiang Bawei Powder have certain antidepressive effect on mice,which may be related to its regulation effect on monoamine neurotransmitter content.
