Optimization of the Processing Endpoint of Scutellariae Radix Carbonisatum Based on the Correlation Analysis of Morphological,Chemical and Biological Activities

摘  要:目的:基于抗炎药效挖掘黄芩炭炮制过程的形性、化学、生物指标变化规律及其内在关联,寻找黄芩炭炮制终点判断的客观量化指标。方法:改变加热温度或时间以制备不同炮制程度的黄芩炭,使用色差仪测定饮片粉末色度值(L^(*)、a^(*)、b^(*))作为形性指标;HPLC测定浸膏中黄芩苷、汉黄芩苷、黄芩素、汉黄芩素的含量作为化学指标;以黄芩炭浸膏对LPS诱导的Raw264.7细胞释放NO、IL-6的抑制率作为生物指标。通过多元统计方法分析三类指标之间的关联性。结果:随着加热程度的增加,黄芩炭饮片颜色逐渐加深,L^(*)、b^(*)持续降低,a^(*)先增后减;黄芩炭饮片及其浸膏中的黄芩苷、汉黄芩苷持续减少,黄芩素、汉黄芩素先增后减;黄芩炭浸膏对IL-6的抑制率先增后减,过度加热时对NO的抑制率明显降低。相关性分析与主成分分析显示,黄芩炭饮片的L^(*)、b^(*)、黄芩苷、汉黄芩苷之间具有正相关关系,a^(*)、黄芩素、汉黄芩素之间具有正相关关系,NO抑制率、IL-6抑制率与前述两组指标均具有一定的相关性。综合三性指标结果,150℃炒制50 min、150℃炒制60 min、180℃炒制30 min、210℃炒制20 min的黄芩炭炮制适中。结论:黄芩炭炮制过程的形性、化学、生物指标具有明显的变化规律与相互关联。根据细胞抗炎药效,黄芩素、汉黄芩素含量达到极大值时为黄芩炭炮制终点,而此终点可以通过a^(*)达到极大值进行快速判断。Objective:Based on the anti-inflammatory activity,the change rule and internal correlation of the triple-indicators of"morphological-chemical-biological"during the processing of Scutellariae Radix carbonisatum were explored to find the objective and quantitative indexes for the processing endpoint.Methods:Scutellariae Radix scorched to different degrees were prepared by changing the heating temperature and time.The color values(L^(*),a^(*),b^(*))of the powdered decoction pieces were determined by a spectrophotometer and used as the morphological indicators.The contents of baicalin,wogonoside,baicalein and wogonin in the decoction water extracts were determined by HPLC and used as the chemical indicators.The inhibition rates of the water extracts on the release of NO and IL-6 from the LPS-induced Raw264.7 cells were used as the biological indicators.Multivariate statistical methods were used to analyze the correlation among the three kinds of indicators.Results:As the increase of the heating degree,the color of the decoction pieces became darker.L^(*)and b^(*)continued to decrease,while a^(*)increased at first but decreased later.The contents of baicalin and wogonoside in the decoction pieces and water extracts decreased continuously,while the contents of baicalein and wogonin increased at first but then decreased.The inhibition rate of IL-6 increased at first but then decreased,while the inhibition rate of NO decreased significantly when overheated.Correlation analysis and principal component analysis showed that L^(*),b^(*),baicalin and wogonoside were significantly positively correlated with each other in Scutellariae Radix carbonisatum decoction pieces.Meanwhile,a^(*),baicalein and wogonin were significantly positively correlated with each other in Scutellariae Radix carbonisatum decoction pieces.The inhibition rates of NO and IL-6 were correlated with both of the above two groups of indicators in Scutellariae Radix carbonisatum decoction pieces.According to the results of the triple-indicators,it was consider
