摘  要:目的:通过建立小鼠T1DN模型,观察对照组、T1DN组及PDTC组的实验结果,研究吡咯烷二硫氨基甲酸酯(PDTC)对血管内皮生长因子(VEGF),色素上皮衍生因子(PEDF)二者表达的影响,探讨PDTC在糖尿病早期使用对糖尿病肾病(DN)的发展影响。方法:36只SPF级雄性BALB/c小鼠,适应性喂养后随机分为对照组,T1DN组及PDTC组,每组12只,将STZ用柠檬酸缓冲液配制为20mg/mL的STZ注射液,以0.1mL/10g对T1DN组、PDTC组小鼠进行一次性腹腔注射制作1型糖尿病模型,对照组给予盐水注射,成膜后(空腹血糖≥16.7mmol/L,C肽≤0.8ng/mL),PDTC组腹腔注射PDTC 60mg/kg隔日一次,持续2周,另外两组同体积生理盐水腹腔注射。T1DN组、PDTC组连续4周出现尿微量白蛋白,C肽≤0.8ng/mL,考虑1型糖尿病肾病成膜。小鼠记录体质量后麻醉,拔眼球采血分离血清检测BUN,Cr、HbA1c,FINS,随后处死并观察肾脏,胰腺病理改变,免疫组化检测VEGF、PEDF表达水平。结果:T1DN组、PDTC组小鼠BUN,Cr、FINS及β细胞功能指数降低,HbAlc增高(P<0.05);与T1DN组相比,PDTC组小鼠BUN,Cr、FINS及β细胞功能指数升高,HbAlc降低(P<0.05)。T1DN组小鼠肾组织中VEGF的表达增高,PEDF的表达降低;PDTC组小鼠肾组织中VEGF的表达降低,PEDF的表达增高。光镜下T1DN组胰腺萎缩,胰岛数量明显减少,伴胰岛萎缩,体积变小,肾小管上皮细胞肿胀,胞浆疏松淡染,肾小管上皮细胞核肿大,肾小球毛细血管丛清晰,PDTC组病理改变轻于T1DN组。结论:PDTC在糖尿病肾病小鼠中下调VEGF、上调PEDF的表达,对1型糖尿病肾病有保护作用。Objective:To discuss the effect of pyrrolidinedithiocarbamic acid ammonium salt(PDTC)on the expression of vascular endothelial growth factor(VEGF)and pigment epithelium-derived factor(PEDF)by establishing mice T1DN model and observing the experimental results of control group,T1DN group and PDTC group,and to explore the effect of PDTC on the development of diabetes nephropathy(DN).Method:Thirty-six SPF male BABL/c mice were randomly divided into control group,T1DN group and PDTC group after adaptive feeding,with 12 mice in each group.STZ was prepared into 20mg/mL STZ injection with citric acid buffer solution,and 0.1mL/10g STZ was intraperitoneally injected once to T1DN group and the PDTC group mice to create Type1 diabetes model.The control group was injected with saline.After membrane formation(fasting blood glucose≥16.7mmol/L,C-peptide≤0.8 ng/mL),PDTC group was intraperitoneally injected with PDTC 60mg/kg every other day,continue for 2 weeks,and the other two groups were intrapertioneally injected with the same volume of physiological saline.In T1DN group and PDTC group,urinary microalbumin with C-peptide≤0.8ng/mL occurred continuously for 4weeks,and membranous nephropathy of Type1 diabetic was considered.The mice were anesthetized after recording their weight,and their eyeballs were removed to extract bllood and isolate serum for detection of BUN,Cr,HbA1c,FINS,and the mice were killed to observe the pathological changes in the kidneys and pancreas.The expression levels of VEGF and PEDF were detected by immunohistochemistry.Results:BUN,Cr,FINS,andβcell function index decreased and HbAlc increased(P<0.05);Compared with the T1DN group,the PDTC group mice showed BUN,Cr,FINS,and theβcell function index increased and HbAlc decreased(P<0.05).The expression of VEGF increased and the expression of PEDF decreased in the renal tissue of T1ND group mice;The expression of VEGF in the kidney tissue of PDTC group mice decreased,while the expression of PEDF increased.Under light microscopy,pancreatic atrophy was obse
