黄芪甲苷调节Apelin/APJ轴抑制喉癌Hep-2细胞的机制

摘　　要：目的探讨黄芪甲苷对喉癌Hep-2细胞增殖、凋亡、迁移和侵袭的影响以及对Apelin/APJ信号轴的调节作用。方法不同浓度(0、20、40、80μmol·L^(-1))黄芪甲苷溶液处理第3代对数生长期Hep-2细胞,检测细胞存活率、凋亡率、迁移和侵袭量细胞数量以及Apelin和APJ蛋白的相对表达量。结果随黄芪甲苷浓度的增加,细胞存活率逐渐降低,细胞凋亡率逐渐升高,迁移和侵袭细胞数量逐渐减少,Apelin和APJ蛋白的相对表达量逐渐降低,且具有浓度依赖性(P<0.05)。结论黄芪甲苷可降低喉癌Hep-2细胞增殖、迁移和侵袭能力,诱导癌细胞凋亡,其可能是通过抑制Apelin/APJ信号通路发挥作用。Objective To investigate the effects of astragaloside on the proliferation,apoptosis,migration and invasion of laryngeal carcinoma Hep-2 cells and the regulation of Apelin/APJ signaling axis.Methods The 3rd passage Hep-2 cells were treated with different concentrations of astragaloside(0,20,40 and 80μmol·L^(-1)).The cell survival rate,apoptosis rate,number of migrating and invading cells,Apelin and APJ protein relative expression levels were measured.Results With the increase of astragaloside concentration,the cell survival rate was gradually decreased,the cell apoptosis rate was increased,the number of migrating and invading cells was gradually decreased,and the relative expression levels of Apelin and APJ proteins were gradually decreased in a concentration dependent manner(P<0.05).Conclusion Astragaloside can reduce the proliferation,migration and invasion of laryngeal carcinoma Hep-2 cells,and induce apoptosis of cancer cells,possibly by inhibiting the Apelin/APJ signaling pathway.
