摘 要:本研究在单因素试验基础进行正交试验,采用Cell Counting Kit-8评价顺铂、二甲双胍和槲皮素不同剂量联合用药方案对鼻咽癌细胞增殖的作用,利用SPSS20.0软件对正交试验结果进行方差和组间差异显著性SSR检验,优选三药联用最佳组合方案。结果表明,随着顺铂、二甲双胍和槲皮素浓度的升高,其对Sune-1细胞的抑制作用越显著,这三种药物对Sune-1的IC_(50)值分别为30.68、6.85、81.96μmol/L,其中顺铂对Sune-1细胞的IC_(50)值最低,表明Sune-1细胞对顺铂的敏感程度强于其他两种药物,槲皮素次之;三药联用有强大抗肿瘤活性,三者的贡献水平由高到低分别为顺铂、二甲双胍、槲皮素,当顺铂浓度61.36μg/mL、二甲双胍浓度13.71 mmol/L、槲皮素浓度163.92μmol/L的高剂量联合用药方案时对人鼻咽癌Sune-1细胞增殖的抑制作用最强,但是,中、低剂量的联合用药方案对人鼻咽癌Sune-1细胞增殖的抑制率也都超过50%,因此,临床上可结合药物毒副作用、患者耐受程度、经济情况等综合因素,考虑选用较低剂量的联合用药方案,为后续临床研究中三药联合的最佳剂量分配提供理论基础。三药联用将成为鼻咽癌的高效化疗方案。In this study,an orthogonal test was carried out on the basis of a single factor test.Cell Counting Kit⁃8 was used to eva⁃luate the effect of different doses of combination medication by Cisplatin,Metformin and Quercetin on the proliferation of nasopharyn⁃geal carcinoma cells,using SPSS20.0 software to carry out SSR test of variance and significant difference between groups on orthogo⁃nal test to select the best combination regimens of the three drugs.The results showed that with the increasing of the concentrations of three drugs about Cisplatin、Metformin and Quercetin,the inhibitory effection about Sune⁃1 cells became more significant.The IC_(50)values of these three drugs on Sune⁃1 cells were 30.68μg/mL,6.855 mmol/L and 81.96μmol/L,respectively,and the Cisplatin IC_(50)values of Sune⁃1 cells were the lowest.The results also showed that the infection of Cisplatin on Sune⁃1 cells was more sensitive than the other two drugs,and the quercetin was followed.When three drugs are used together,the combination showed the strong anti⁃tumor activity,and the contribution levels of the three drugs from high to low were Cisplatin,Metformin and Quercetin,respectively.With the high⁃dose combination regimen of Cisplatin concentration of 61.36μg/mL,Metformin concentration of 13.71 mmol/L and Quercetin concentration of 163.92μmol/L,there was the strongest inhibitory effection on the proliferation of the human nasopharyn⁃geal carcinoma Sune⁃1 cells,however,the combination regimens of medium or low doses also inhibited more than 50%the prolifera⁃tion of human nasopharyngeal carcinoma Sune⁃1 cells.Therefore,the drug toxicity effects、patient tolerance、economic conditions and other comprehensive factors could be considered to select a lower⁃dose combination of drug regimen,providing a theoretical basis for the optimal dose allocation of the combination of three drugs in subsequent clinical studies.The combination of three drugs about Cisplatin,Metformin and Quercetin would be an effective chemo
