摘 要:目的分析某铁路辖区列车水箱水中细菌种群多样性。方法采集某地区始发列车水箱水,用营养琼脂平板分菌,根据菌落形态、革兰染色对生长的全部细菌进行分类、计数,并进一步鉴定到种属。分离出的细菌采用16S测序,通过NCBI数据库比对得到种属结果;同时采用生化试验将细菌鉴定到种属。结果列车水箱水共检出22个菌属,其中普车与动车共有9个菌属,普车特有6个菌属,动车特有7个菌属。普车与动车检出细菌相对丰度与频度最高的菌属均为假单胞菌属。结论普车与动车水箱水样本细菌菌群分布存在差异性;水箱生境存在一定程度的假单胞菌蓄积。Objective This paper aims to analyze the diversity of bacterial populations in the tank water of trains in a railroad jurisdiction.Methods The water in the tank of a regional starting train was collected and separated by nutrient agar plate,and the bacteria were separated by nutrient AGAR plate.All the growing bacteria were classified and counted according to colony morphology and gram staining,and the species were further identified.The isolated bacteria were sequenced by 16S,and the species results were obtained by NCBI database comparison.At the same time,the bacterial species were identified by biochem⁃ical tests.Results A total of 22 genera were detected,including 9 genera in both regular trains and high-speed trains,6 gen⁃era only in regular trains,and 7 genera only in high-speed trains.Pseudomonas was the genus with the highest relative abun⁃dance and frequency of bacteria detected in both regular trains and high-speed trains.Conclusion There are differences in the distribution of bacterial flora between the tank water samples of regular trains and bullet/high-speed trains.There was a certain degree of accumulation of Pseudomonas in the tank habitat.
