摘  要:植物病原真菌分泌蛋白在其致病过程中起着重要的作用。通过NCBI数据库下载前人已报道的24种炭疽属植物病原真菌全基因组序列,利用SignalP v5.0、ProtComp v9.0、TMHMM v2.0等在线分析程序对这些炭疽菌蛋白质的跨膜结构、信号肽、糖基磷脂酰肌醇(GPI)锚定位点、亚细胞定位等进行预测分析,根据分泌蛋白的典型特征筛选获取24种炭疽菌的分泌蛋白。结果显示:在约34万条蛋白序列中,分泌蛋白所占比例为2.14%~3.76%,其氨基酸残基数量多集中于100~300个,并且氨基酸组成相似度较高,其中以丙氨酸残基在分泌蛋白中的含量最高,所占比例达9.72%~10.54%;同时,24种炭疽菌分泌蛋白的信号肽在-3和-1位切割位点处相对比较保守,属于A-X-A类型,其氨基酸残基数量多集中于17~22个。可见,炭疽属不同种之间分泌蛋白的比例、氨基酸组成、信号肽特征等存在差别,但差异并不大。Secreted proteins in plant pathogenic fungi play an important role in the pathogenic process.In this study,based on the whole genome sequences of 24 species of Colletotrichum reported by predecessors,the transmembrane structure,signal peptide,GPI anchor point and subcellular localization of 24 Colletotrichum protein were predicted and analyzed by on-line analysis programs such as SignalP v5.0,ProtComp v9.0 and TMHMM v2.0,respectively.And 24 secreted proteins of Colletotrichum were obtained according to the typical characteristics of secreted proteins.The results show that among about 340000 protein sequences,the proportion of secreted proteins is 2.14%-3.76%,the amino acid length is mostly concentrated in 100-300,and the composition of amino acid has high similarity.The content of alanine residues in secreted proteins is the highest,accounting for 9.72%-10.54%.The signal peptides of 24 fungi secreted proteins are relatively conservative at the cleavage sites of-3 and-1,belonging to A-X-A type,and their amino acid lengths are mostly concentrated in 17-22.There are differences in the proportion of secreted proteins,amino acid composition and signal peptide characteristics among different species of Colletotrichum,but the differences are not big.
