摘 要:旨在研究灌服丁酸钠对哺乳羔羊瘤胃内容物和瘤胃上皮微生物菌群区系的影响。试验选取7对体重相近、胎次一致的10日龄湖羊双胎羔羊,随机分为2组,一组为口腔灌服丁酸钠组(试验组,OB,n=7),另一组为灌服生理盐水组(对照组,CON,n=7)。羔羊在49日龄时屠宰采样,采取瘤胃内容物和瘤胃上皮样品,分别用于挥发性脂肪酸(VFA)及16S rRNA高通量测序分析。结果显示,口腔灌服丁酸钠显著影响了瘤胃内丁酸和总VFA的含量(P<0.05),但并未影响乙酸、丙酸的含量以及乙酸丙酸比值(乙丙比)(P>0.05)。冗余分析(RDA)结果表明,对照组和试验组之间瘤胃上皮微生物菌群结构差异较大,而瘤胃内容物微生物区系的差异较小。对瘤胃内容物微生物来说,灌服丁酸钠显著提高了BS11肠道菌族(Unclassified BS11 gut group)的丰度(P<0.05),显著降低了梭菌目(Unclassified Clostridiales)和丹毒丝菌科(Unclassified Erysipelotrichaceae)的丰度(P<0.05);对瘤胃上皮微生物来说,灌服丁酸钠显著了提高拟杆菌目(Unclassified Bacteroidales)、密螺旋体(Treponema)的相对丰度(P<0.05),显著降低了弯曲杆菌属(Campylobacter)、普雷沃氏菌科(Unclassified Prevotellaceae)、拟杆菌属(Bacteroides)、γ-变形杆纲(Unclassified Gammaproteobacteria)、梭菌目和月形单胞菌属(Selenomonas)的相对丰度(P<0.05)。结果表明,口腔灌服丁酸钠可显著改变哺乳羔羊瘤胃上皮微生物菌群结构,但对瘤胃内容物微生物区系和瘤胃发酵参数的影响较小。To determine the effect of sodium butyrate on the rumen fermentation and bacterial community of rumen contents and rumen epi⁃thelial in sucking lambs,seven pairs of 10-day-old twin Hu lambs were obtained and randomly assigned to either an oral administration of sodium butyrate group(OB,n=7)or a physiological saline group(CON,n=7).The results showed that oral administration of sodium buty⁃rate had a significant effect on the concentration of butyrate and total VFA in the rumen of the lambs(P<0.05),but not on the concentration of acetate,propionate,and the acetate/propionate ratio(P>0.05)therein.The results of the redundancy analysis(RDA)showed a greater difference in the bacterial structure of rumen epithelium between the OB and CON groups,compared with the bacterial community of rumen contents which showed little difference.For rumen content microbiota,the relative abundance of the Unclassified BS11 gut group was signifi⁃cant higher in the lambs of the OB group than that in the CON group(P<0.05).Compared with the OB group,the CON group had higher relative abundance of Unclassified Clostridiales and Unclassified Erysipelotrichaceae(P<0.05).For rumen epithelial microbiota,the relative abundance of Unclassified Bacteroidales and Treponema were significantly higher in the lambs of the OB group than those in the CON group(P<0.05).And the relative abundance of Campylobacter,Unclassified Prevotellaceae,Bacteroides,Unclassified Gammaproteobacteria,Unclassified Clostriales and Selenomonas was higher in the CON group than in the OB group(P<0.05).These results suggested that oral ad⁃ministration of sodium butyrate produced significant effect on the bacterial structure in the rumen epithelium of suckling lambs but had limited effect on rumen content bacterial community and rumen fermentation parameters.
